Description:
The 1A8 monoclonal antibody specifically reacts with the 21-25 kDa glycophosphatidylinositol-anchored protein known as Ly-6G, expressed by the granulocytes from the bone marrow and periphery neutrophils. Ly-6G and Ly-6C form the Granulocyte Receptor-1 antigen (GR-1). The binding of the 1A8 antibody to the Ly-6G can be blocked by another antibody, RB6-8C5, which also recognizes Ly-6C. While 1A8 is specific for only Ly-6G, RB6-8C5 also binds to Ly-6C.

Description:
The 30-H12 monoclonal antibody specifically binds to mouse CD90.2, an alloantigen known as Thy-1.2, expressed on thymocytes, mature T cells, epithelial cells, neurons, hematopoietic stem cells, and fibroblasts. CD90 is a membrane glycoprotein that regulates the adhesion and signal transduction in T lymphocytes, and the adhesion of thymocytes to thymic stroma. The interaction between 30-H12 and the antibody to the CD3/TCR complex upregulates thymocytes signal transduction and apoptosis and downregulates mature T cell proliferation. The 30-H12 antibody seems to be unable to cross-link with CD90.1.

Description:
Tau is the principal microtubule-associated protein located in axons in the nervous system where it participates in events including the elongation, stabilisation and bundling of microtubules. The tau family consists of six isoforms identified by the insertion of three or four partial repeats of 30-31 amino acids in the C-terminal region. Subcellular fractionation studies have revealed tau to be associated with the membrane fraction, as well as with the cytoskeletal fraction and there is some evidence that the N-terminal domain of tau may be responsible for membrane binding as well as interaction with other cellular components. Tau extracted from human fetal and neonatal brains is moderately phosphorylated with 5-9 phosphorylated serine/threonine residues per molecule. Normal adult human brain tau shows a marked reduction in phosphorylation, but in certain neurodegenerative diseases (tauopathies) tau becomes hyperphosphorylated (6-15 phosphates per tau molecule have been identified) and associates to form filamentous aggregates. In Alzheimer’s disease (AD) hyperphosphorylated tau polymerises to form a mixture of paired helical filaments (PHF) and straight filaments which accumulate and contribute to the formation of neurofibrillary tangles - one of the characteristic features of AD. It was reported in 1998 that tau could be tyrosine phosphorylated when co-transfected with the src-family protein tyrosine kinase fyn. In 2004 the same group, using phospho-specific antibodies, reported that fyn phosphorylated Tyr18 of brain tau at an early developmental stage in mice, but that tyrosine phosphorylated tau was undetectable in adult mice. However, they also showed that PHF preparations were reactive to the tau-pTyr18-specific antibodies, implicating a role for fyn – and possibly other tyrosine kinases - in the neurodegenerative process.

Description:
Serine/threonine protein kinase which is a central regulator of cellular metabolism, growth and survival in response to hormones, growth factors, nutrients, energy and stress signals. MTOR directly or indirectly regulates the phosphorylation of at least 800 proteins. Functions as part of 2 structurally and functionally distinct signaling complexes mTORC1 and mTORC2 (mTOR complex 1 and 2). Activated mTORC1 up-regulates protein synthesis by phosphorylating key regulators of mRNA translation and ribosome synthesis. This includes phosphorylation of EIF4EBP1 and release of its inhibition toward the elongation initiation factor 4E (eiF4E). Moreover, phosphorylates and activates RPS6KB1 and RPS6KB2 that promote protein synthesis by modulating the activity of their downstream targets including ribosomal protein S6, eukaryotic translation initiation factor EIF4B, and the inhibitor of translation initiation PDCD4. Stimulates the pyrimidine biosynthesis pathway, both by acute regulation through RPS6KB1-mediated phosphorylation of the biosynthetic enzyme CAD, and delayed regulation, through transcriptional enhancement of the pentose phosphate pathway which produces 5-phosphoribosyl-1-pyrophosphate (PRPP), an allosteric activator of CAD at a later step in synthesis, this function is dependent on the mTORC1 complex. Regulates ribosome synthesis by activating RNA polymerase III-dependent transcription through phosphorylation and inhibition of MAF1 an RNA polymerase III-repressor. In parallel to protein synthesis, also regulates lipid synthesis through SREBF1/SREBP1 and LPIN1. To maintain energy homeostasis mTORC1 may also regulate mitochondrial biogenesis through regulation of PPARGC1A. mTORC1 also negatively regulates autophagy through phosphorylation of ULK1. Under nutrient sufficiency, phosphorylates ULK1 at 'Ser-758', disrupting the interaction with AMPK and preventing activation of ULK1.

The J116 monoclonal antibody specifically reacts with human Programmed death-1 (PD-1 or CD279), a 50-55 kDA glycoprotein. It is expressed on mainly on activated B, T, and myeloid cells. Within the cytoplasmic region, PD-1 contains an Immunoreceptor tyrosine-based inhibitory motif (ITIM) and seems to regulate peripheral tolerance. The lack or mutation of CD279 is linked to autoimmune disorders. The J116 antibody does not block the binding of the PD-1 ligand, but does inhibit the PD-1 signal transduction.

The 17A2 monoclonal antibody specifically reacts with the mouse T lymphocytes receptor (TCR) associated CD3 complex, resulting in cellular activation and proliferation. CD3 is expressed by thymocytes and mature lymphocytes, and contains γ, δ, and ε subunits, involved in the assembly, trafficking, and surface expression of T-cell receptor complex. The interaction between the T lymphocytes and the 17A2 antibody can be blocked by the 145-2C11 anti-CD3e antibody, demonstrating that the 17A2 antibody recognizes the CD3 ε chain.

Description:
Product is the lyophilized powder of goat antiserum to human secretory IgA and buffer salts.
IgA antibody is a secretory antibody found abundantly in mucous linings of gastrointestinal, respiratory and genitourinary tracts, tears and saliva. Secretory IgA contributes to the humoral defense mechanism against the pathogens on mucosal surfaces. Anti-Human IgA (α-chain specific) antibody specifically recognizes human IgA. Immunoglobulin A (IgA) is the primary antibody involved in mucosal immunity. It is identified by the presence of α-type heavy chains. IgA, which is composed of two subclasses IgA1 and IgA2, can exist in a dimeric secretory form found in mucous secretions. The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer is standardized by equivalence-point precipitation. Each antiserum is tested for specificity at a minimum of 80 mg/mL using immunoelectrophoresis. The antiserum is prepared by hyperimmunizing goats with highly purified human secretory IgA; antiserum is delipidated and made specific by solid-phase absorption. The resulting antiserum is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, with 0.05% sodium azide, adjusted to standard titer, filtered through a 0.22 µm filter, vialed and lyophilized. This product is suitable for use as a precipitating antibody. It gives sharp precipitin lines in immunoelectrophoresis and immunodiffusion techniques. It is suitable for Ouchterlony double diffusion. The antibody was used for IgA quantification by ELISA in various studies. Antibody dilution of 1:4000 was used for passive hemagglutination inhibition assay.
