Folic acid, also known as folate, is a B vitamin that can be found in a variety of fruits and vegetables. It can also be chemically synthesized. Folate, a watersoluble vitamin, helps the body form red blood cells and aids in the formation of genetic material within every body cell. This product exhibits metal binding properties. Hematopoietic vitamin present, free or combined with one or more additional molecules of L-(+)-glutamic acid, in liver, kidney, mushrooms, spinach, yeast, green leaves, and grasses.
Folic acid (Vitamin B9 and folate) is essential to numerous bodily functions. The human body needs folate to synthesize DNA, repair DNA, and methylate DNA as well as to act as a cofactor in certain biological reactions. It is especially important in aiding rapid cell division and growth, such as in infancy and pregnancy. Folic acid (FA) and dihydrofolic acid (FAH2) are substrates of dihydrofolate reductase(s) which reduce them to tetrahydrofolate (THF), which in turn supports ‘one carbon’ transfer. Tetrahydrofolates are required for de novo synthesis of purines, thymidylic acid and various amino acids and for post-translational methylation (epigenetics). Very slightly soluble in cold water (0.0016 mg/mL at 25 °C), soluble to about 1% in boiling water. Slightly soluble in methanol, appreciably less soluble in ethanol and butanol. Insoluble in acetone, chloroform, ether, benzene. Relatively soluble in acetic acid, phenol, pyridine, solutions of alkali hydroxides and carbonates. Soluble in hot dilute HCl and H2SO4.

The Hoechst stains are a family of fluorescent stains for labeling DNA in fluorescence microscopy. The blue fluorescent Hoechst dye is a cell permeable nucleic acid stain that has multiple applications, including sensitive detection of DNA in the presence of RNA in agarose gels, automated DNA determination, sensitive determination of cell number and chromosome sorting. Useful vital stain for the flow cytometric recognition of DNA damage and other viability measurements by monitoring the emission spectral shifts of the dyes. Because this fluorescent stain labels DNA, it can also be used to visualize nuclei and mitochondria. Hoechst 34580 can be excited by violet laser at 405 nm, and emit blue/cyan fluorescence light around an emission maximum at ~440 nm. It is cell-permeable and an excellent tool for flow cytometry studies.

The lymphocyte-specific kinase (Lck) is a cytoplasmic tyrosine kinase of the Src family expressed in T cells and natural killer (NK) cells. Genetic evidence in both mice and humans demonstrates that Lck kinase activity is critical for signaling mediated by the T cell receptor (TCR), which leads to normal T cell development and activation. Selective inhibition of Lck is expected to offer a new therapy for the treatment of T-cell-mediated autoimmune and inflammatory disease. AMG-47 is one of two optimized chemical compounds that has been shown to be extremely effective in vivo and in vitro in inhibiting Lck as well as a number of other receptor tyrosine kinases. In an anti-CD3/ IL-2 mouse model system AMG-47 has been shown to be effective in inhibiting the Lck mediated anti-inflammatory activity (ED50 11 mg/kg; 630nM) in vivo. In multiple other in vitro assays, AMG-47 exhibits subnanomolar inhibition against Lck, and low (<lt/>10nM) inhibition against other hard to inhibit kinases such as KDR and SRC and MAPK alpha (p38alpha). Moreover at slightly higher doses but well under 10µM, AMG-47 effectively inhibits the JNK family of kinases including TYK2 at ~ 1.2µM.

Thermo Scientific Pierce SMCC is a hetero-bifunctional crosslinker that contain N-hydroxysuccinimide (NHS) ester and maleimide groups that allow covalent conjugation of amine- and sulfhydryl-containing molecules. NHS esters react with primary amines at pH 7–9 to form amide bonds, while maleimides react with sulfhydryl groups at pH 6.5–7.5 to form stable thioether bonds. In aqueous solutions, NHS ester hydrolytic degradation is a competing reaction whose rate increases with pH. The maleimide group is more stable than the NHS-ester group, but will slowly hydrolyze and lose its reaction specificity for sulfhydryls at pH values > 7.5. For these reasons, conjugations with these crosslinkers are usually performed at pH 7.2–7.5, with the NHS ester (amine-targeted) reacted before or simultaneous with the maleimide (sulfhydryl-targeted) reaction.

SAR131675 is a potent and selective VEGFR-3 inhibitor. It inhibited VEGFR-3 tyrosine kinase activity and VEGFR-3 autophosphorylation in HEK cells with IC(50) values of 20 and 45nM, respectively. SAR131675 dose dependently inhibited the proliferation of primary human lymphatic cells, induced by the VEGFR-3 ligands VEGFC and VEGFD, with an IC(50) of about 20nM. SAR131675 was found to be highly selective for VEGFR-3 versus 107 receptors, enzymes, ion channels, and 65 kinases. However, it was moderately active on VEGFR-2 with a VEGFR-3/VEGFR-2 ratio of about 10. SAR131675 had no antiproliferative activity on a panel of 30 tumors and primary cells, further showing its high specificity and indicating that SAR131675 is not a cytotoxic or cytostatic agent.
