Initiation of transcription by RNA polymerase II requires the activities of more than 70 polypeptides. The protein that coordinates these activities is the basal transcription factor TFIID, which binds to the core promoter to position the polymerase properly, serves as the scaffold for assembly of the remainder of the transcription complex, and acts as a channel for regulatory signals. TFIID is composed of the TATA-binding protein (TBP) and a group of evolutionarily conserved proteins known as TBP-associated factors or TAFs. TAFs may participate in basal transcription, serve as coactivators, function in promoter recognition or modify general transcription factors (GTFs) to facilitate complex assembly and transcription initiation. This gene encodes the largest subunit of TFIID. This subunit binds to core promoter sequences encompassing the transcription start site. It also binds to activators and other transcriptional regulators, and these interactions affect the rate of transcription initiation. This subunit contains two independent protein kinase domains at the N and C-terminals, but also possesses acetyltransferase activity and can act as a ubiquitin-activating/conjugating enzyme. This gene is part of a complex transcriptional unit (TAF1/DYT3), wherein some products share exons with TAF1 as well as additional exons downstream.

Description:
Gonadotropin-releasing hormone (GnRH) is released in a pulsatile manner that varies with the reproductive cycle. This hypothalamic hormone is transported to the pituitary, where it binds to specific receptors and regulates the synthesis and release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Localized to the cell membrane, GnRHR2 mediates its own receptor activity via association with G proteins, thereby activating a phosphatidylinositol-calcium second messenger system that regulates GnRHR2 function. GnRHR2 is thought to have potent antiproliferative effects on ovarian and endometrial cancer cells, suggesting a possible role in tumor suppression. Due to alternative splicing events, GnRHR2 is expressed as two isoforms.

The KV gene family encodes more than 30 genes that comprise the subunits of the K+ channels, and they vary in their gating and permeation properties, subcellular distribution, and expression patterns. Functional KV channels assemble as tetramers consisting of pore-forming alpha-subunits (KV alpha), which include the KV1, KV2, KV3, and KV4 proteins, and accessory or KV beta subunits that modify the gating properties of the coexpressed KV alpha subunits. Differences exist in the patterns of trafficking, biosynthetic processing and surface expression of the major KV1 subunits (KV1.1, KV1.2, KV1.4, KV1.5 and KV1.6) expressed in rat and human brain, suggesting that the individual protein subunits are highly regulated to control for the assembly and formation of functional neuronal channels.
