Description:
Polycomb group (PcG) protein. Catalytic subunit of the PRC2/EED-EZH1 complex, which methylates 'Lys-27' of histone H3, leading to transcriptional repression of the affected target gene. Able to mono-, di- and trimethylate 'Lys-27' of histone H3 to form H3K27me1, H3K27me2 and H3K27me3, respectively. Required for embryonic stem cell derivation and self-renewal, suggesting that it is involved in safeguarding embryonic stem cell identity. Compared to EZH1-containing complexes, it is less abundant in embryonic stem cells, has weak methyltransferase activity and plays a less critical role in forming H3K27me3, which is required for embryonic stem cell identity and proper differentiation. Sequence similarities Belongs to the histone-lysine methyltransferase family. EZ subfamily.Contains 1 SET domain.

Description:
Acetylation and methylation of lysine are important post-translational modifications that regulate numerous protein-protein and protein-DNA interactions. Lysine acetylation and methylation involves the transfer of acetylCoA, or one or more methyl groups, to the e-amino group of lysine by modifying enzymes and cofactors. Histones and transcription factors are the primary targets of lysine acetylation and methylation, with either modification capable of inducing gene silencing or expression due to differential regulation of cofactors. For example, varying degrees of mono-, di-, and tri-methylation or acetylation of histone H3 at lysine residue 9 are known to demark distinct chromatin regions during various states of gene activation (methylation) or repression (acetylation).

Description:
Acts both as NAD-dependent protein ADP-ribosyl transferase and NAD-dependent protein deacetylase. Catalyzes the transfer of ADP-ribosyl groups onto target proteins, including mitochondrial GLUD1, inhibiting GLUD1 enzyme activity. Acts as a negative regulator of mitochondrial glutamine metabolism by mediating mono ADP-ribosylation of GLUD1: expressed in response to DNA damage and negatively regulates anaplerosis by inhibiting GLUD1, leading to block metabolism of glutamine into tricarboxylic acid cycle and promoting cell cycle arrest. In response to mTORC1 signal, SIRT4 expression is repressed, promoting anaplerosis and cell proliferation. Acts as a tumor suppressor. Also acts as a NAD-dependent protein deacetylase: mediates deacetylation of 'Lys-471' of MLYCD, inhibiting its activity, thereby acting as a regulator of lipid homeostasis. Down-regulates insulin secretion.
