Description:
The protein encoded by this gene, a member of the peptidase C1 family, is a lysosomal cysteine proteinase that appears to be a central coordinator for activation of many serine proteinases in immune/inflammatory cells. It is composed of a dimer of disulfide-linked heavy and light chains, both produced from a single protein precursor, and a residual portion of the propeptide acts as an intramolecular chaperone for the folding and stabilization of the mature enzyme. This enzyme requires chloride ions for activity and can degrade glucagon. Defects in the encoded protein have been shown to be a cause of Papillon-Lefevre syndrome, an autosomal recessive disorder characterized by palmoplantar keratosis and periodontitis. Gamma-Aminobutyric Acid Receptor-Associated Protein (GABARAP) is a ligand-gated chloride channel protein that mediates inhibitory neurotransmission and belongs to the MAP1 LC3 family. GABARAP is highly positively charged in its N-terminus and shares sequence similarity with light chain-3 of microtubule-associated proteins 1A and 1B. GABARAP clusters neurotransmitter receptors by mediating interaction with the cytoskeleton. Autophagy is the process by which cells recycle cytoplasm and dispose of excess or defective organelles. This process is suggested to be involved development, differentiation, growth regulation and tissue remodeling in multicellular organisms. An important inhibitory neurotransmitter, GABA, acts on GABA receptors that are ubiquitously expressed in the CNS. GABARAP has been shown to play a important role in intracellular transport of GABA(A) receptors and its interaction with the cytoskeleton.

OXSR1 is a serine/threonine kinase which regulates downstream kinases in response to environmental stress such as osmotic stresses, notably sorbitol and, to a lesser extent, NaCl. OXSR1 phosphorylated thr84 within the N-terminal regulatory domain of PAK1. Replacement of thr84 with gln reduced activation of PAK1 by an active form of the small G protein CDC42, suggesting that phosphorylation by OXSR1 modulates the G protein sensitivity of PAK. OXSR1 interacts with chloride channel proteins SLC12A6 isoform 2, SLC12A1 and SLC12A2 but not with SLC12A4 and SLC12A7, possibly establishing sensor/signaling modules that initiate the cellular response to environmental stress. Binds to and phosphorylates RELL1, RELL2 AND RELT. OXSR1 may have a role in regulating the actin cytoskeleton.
