Transmembrane serine/threonine kinase forming with the TGF-beta type I serine/threonine kinase receptor, TGFBR1, the non-promiscuous receptor for the TGF-beta cytokines TGFB1, TGFB2 and TGFB3. Transduces the TGFB1, TGFB2 and TGFB3 signal from the cell surface to the cytoplasm and is thus regulating a plethora of physiological and pathological processes including cell cycle arrest in epithelial and hematopoietic cells, control of mesenchymal cell proliferation and differentiation, wound healing, extracellular matrix production, immunosuppression and carcinogenesis. The formation of the receptor complex composed of 2 TGFBR1 and 2 TGFBR2 molecules symmetrically bound to the cytokine dimer results in the phosphorylation and the activation of TGFRB1 by the constitutively active TGFBR2. Activated TGFBR1 phosphorylates SMAD2 which dissociates from the receptor and interacts with SMAD4. The SMAD2-SMAD4 complex is subsequently translocated to the nucleus where it modulates the transcription of the TGF-beta-regulated genes. This constitutes the canonical SMAD-dependent TGF-beta signaling cascade. Also involved in non-canonical, SMAD-independent TGF-beta signaling pathways.

The H57-597 monoclonal antibody is specific for the beta chain of the mouse T cell Receptor (TCR). The crosslinking induces activation and proliferation of T cells, as a plate-bound or a soluble H57-597, or the plate-bound antibody can induce apoptosis, based on assay conditions. The beta chain of the TCR can combine with the alpha chain of the receptor to produce the alpha-beta TCR, which is expressed by the NKT cells, by the NK1.1+ thymocytes and most of the T cells. The beta chain does not react with the gamma-delta TCR-bearing cells, expressed by a small number of T cells. This antibody can be used as a phenotypic marker for the TCR beta expressing cells or for the functional purpose of TCR-mediated cell activation or apoptosis.

Beta Arrestin 1 is a member of a family of proteins that are widely expressed but especially abundant in the central nervous system. Serving as an adaptor or scaffold molecule, beta Arrestin 1 is essential for mitogenic signaling. It mediates agonist dependent desensitization and internalization of G protein coupled receptors (GPCRs, e.g., beta 2 adrenergic receptor). After binding to their ligand and interacting with heterotrimeric G proteins, GPCRs are phosphorylated by G protein receptor kinases (GRKs) on serine residues. Beta Arrestin 1 has important roles in the cytoplasm and at the plasma membrane in the desensitization and internalization of G protein coupled receptors (GPCRs) and is increasingly appreciated to play an important role in the endocytosis and signaling of GPCRs. Beta Arrestin 1 in the cytosol is phosphorylated by ERK1 and 2 on serine 412 in a negative feedback mechanism and binds to the phosphorylated receptors at the plasma membrane. The Beta-Amyloid precursor is known to interact with several proteins, including X11 and the G heterotrimetric protein APP-BP1. The neuronal, transmembrane protein X11 is known to bind to the ∫-Amyloid precursor protein via a phosphotyrosine binding (PTB) domain, reducing the secretion of cellular Beta-APP and slowing Beta-APP processing pathways. X11 binds specifically to the YENPTY motif, which is involved in the internalization of Beta-APP. Multiple splice varitents of X11 have been identified, including X11 Alpha (also designated Mint 1), X11 Beta (Mint 2) and X11 Gamma(Mint 3).
