Description:
Catalyzes the second step of the reductive pyrimidine degradation, the reversible hydrolytic ring opening of dihydropyrimidines. Can catalyzes the ring opening of 5,6-dihydrouracil to N-carbamyl-alanine and of 5,6-dihydrothymine to N-carbamyl-amino isobutyrate.Tissue specificity:Liver and kidney.Involvement in disease:Defects in DPYS are the cause of dihydropyrimidinase deficiency (DHPD). DHPD is an autosomal recessive disorder characterized by dihydropyrimidinuria and associated with a variable clinical phenotype: epileptic or convulsive attacks, dysmorphic features and severe developmental delay, and congenital microvillous atrophy.

Description:
Potent cell permeable JAK3 inhibitor (IC50 = 1nM). Shows also JAK-1 inhibitory activity. Immunosuppressive and anti-inflammatory agent. Inhibits signaling through heterodimeric receptors associated with JAK3, JAK1, or both of them, with functional selectivity over JAK2-paired receptors. Blocks downstream STAT signaling, resulting in potent inhibition of several cytokines, including interleukins 2, 4, 7, 9, 15 and 21, which are integral to lymphocyte activation, function and proliferation. Investigated for several autoimmune disorders including, rheumatoid arthritis and psoriasis. Potent and selective inhibitor of HIV-1 replication and virus reactivation in vitro.

Description:
Thymidine kinase (TK) belongs to a group of enzymes such as dihydrofolate reductase, thymidylate synthase, and DNA polymerase that are involved in DNA synthesis and precursor production. High levels of these enzymes are present in proliferating cells, and low levels are found in resting cells, due to multiple regulatory mechanisms that ensure exclusive expression of these enzymes in replicating cells. TK is responsible for catalyzing the phosphorylation of thymidine, which functions as a part of the pyrimidine salvage pathway involved in DNA synthesis. The activities of enzymes such as TK are essential for the activation of several chemotherapeutically important nucleoside analogues that are administered as prodrugs.
