The cytokine Tumor necrosis factor-alpha (TNF-alpha) is secreted by activated monocytes/macrophages and T lymphocytes and has been implicated in several disease states, including rheumatoid arthritis, inflammatory bowel disease, septic shock, and osteoporosis. Monocyte/macrophage production of TNF-alpha is dependent on the mitogen-activated protein kinase p38. The compound RWJ 67657 is a potent, orally active inhibitor of MAPK and it inhibited the release of TNF-alpha from LPA stimulated human mononuclear cells with an IC(50) of 3nM, as well as the release of TNF-alpha from peripheral blood mononuclear cells treated with the T cell stimulator and superantigen staphylococcal enterotoxin B with an IC(50) value of 13nM. Also RWJ 67657 was approximately 10-fold more potent than the literature standard p38 kinase inhibitor SB 203580 in all p38 dependent in vitro systems tested and is much more specific than SB 203580 for p38 kinaes. RWJ 67657 inhibited the enzymatic activity of recombinant p38alpha and beta, with in vitro IC(50) values of 1 and 11µM respectively, but not gamma or delta, and in further in vitro test across a panel of other enzymes RWJ 67657 demonstrated no other significant activity.

The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer of the IgG fraction is standardized by equivalence-point precipitation. F(ab')2 fragments are tested for purity and specificity at a minimum of 20 mg/mL using immunoelectrophoresis. The product is goat F(ab')2; no trace of albumin, whole molecule IgG or Fc is detected.
The F(ab')2 fragment is prepared from the specific goat IgG fraction by limited digestion with pepsin, and purified by gel filtration or affinity chromatography. The resulting F(ab')2 fragment is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, 0.05% sodium azide, adjusted to standard titer, filtered through a 0.22 μm filter, vialed and lyophilized. Goat F(AB')2 fragment to Human IgM is used as a precipitating antibody. It gives sharp precipitin lines in immunoelectrophoresis and immunodiffusion techniques. This product is suitable for conjugation with enzymes, radiolabels, fluorochromes or other markers, and for attachment to solid supports for use as an immunoadsorbant. This product is suitable for use as a primary reagent in enzyme immunoassays, fluorescein immunoassays and blot, cell or tissue immunostaining. F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors. (Note: Affinity purified antibodies are recommended to avoid non-specific binding from the inherent antibodies of host animals).

Peroxidase-Conjugated Goat IgG Fraction to Human IgG (Whole Molecule) has been used in dot blot. It is suitable for use as a reagent in enzyme immunoassays (EIA), cell and immunostaining of acetone-fixed frozen and formalin-fixed, paraffin-embedded tissue sections, cell and tissue labeling (for electron microscopy), and blot immunostaining. Note: F(ab')₂ fragments are recommended for staining of cells or tissues which contain Fc receptors. Affinity purified antibodies or their fragments are recommended to avoid non-specific binding from inherent antibodies of host animals. Antibody and highly purified HRP (Rz>3.0) are conjugated under defined conditions to obtain optimally labeled product. Conjugated protein is purified by salt fractionation. The product is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, with 0.01% thimerosal, adjusted to standard titer, filtered through a 0.22 µm filter, vialed and lyophilized. The total protein is measured using the Biuret procedure with bovine albumin as the standard. To ensure that the product titer falls within the required range, antibody titer is standardized by microtiter plate ELISA with human IgG. The product is tested for purity and specificity at final concentration by immunoelectrophoresis.
