Description:
β-NADP is a coenzyme necessary for the alcoholic fermentation of glucose and the oxidative dehydrogenation of other substances. It occurs widely in living tissue, especially in the liver. Nicotinic acid can be converted to nicotinamide in the body and, in this form, is found as a component of two oxidation-reduction coenzymes: nicotinamide adenine dinucleotide (NAD) and nicotinamide adenine dinucleotide phosphate (NADP). The nicotinamide portion of the coenzyme transfers hydrogens by alternating between oxidized quaternary nitrogen and a reduced tertiary nitrogen. NADP is an essential coenzyme for glucose-6-phosphate dehydrogenase which catalyzes the oxidation of glucose-6-phosphate to 6-phosphogluconic acid. This reaction initiates metabolism of glucose by a pathway other than the citric acid cycle. This route is known as the hexose phosphate shunt or phosphogluconate pathway. Other enzymes which utilize NADP as a coenzyme are: Alcohol dehydrogenase:NADP dependent; Aromatic ADH:NADP dependent; Ferredoxin-NADP reductase; L-Fucose dehydrogenase; Gabase; Galactose-1-phosphate uridyl transferase; Glucose dehydrogenase; L-Glutamic dehydrogenase; Glycerol dehydrogenase:NADP specific; Isocitric dehydrogenase; Malic enzymes; 5,10-Methylenetetrahydrofolate dehydrogenase; 6-Phosphogluconate dehydrogenase and Succinic semialdehyde dehydrogenase.

Maelstrom Antibody: The mammalian homolog of the Drosophila protein Maelstrom is expressed in the male germline and localizes to the sex body in spermatocytes and the chromatoid body in round spermatids. Similar to its expression in Drosophila, Maelstrom is a component of nuages, a germ-cell specific organelle and is thought to be essential for spermatogenesis and transposon repression during meiosis. In humans, Maelstrom has been found to be expressed only in the testis and in various cancer cell lines. Treatment of these cell lines with the demethylating agent 5'-Aza-2-Deoxycytidine significantly upregulated Maelstrom levels, indicating that its expression is regulated by DNA methylation.

C1orf77, also known as Friend of PRMT1 protein, is a 248 amino acid protein that plays an essential role in the ligand-dependent activation of estrogen receptor target genes. C1orf77 is tightly associated with chromatin and is modified by both asymmetric and symmetric arginine methylation. Depletion of C1orf77 results in almost complete block of estradiol-induced promter occupancy by the estrogen receptor. Also, complete knockdown of C1orf77 mRNA in adult erythroid progenitors stongly induces fetal hemoglobin, suggesting that C1orf77 is a critical modulator of _-globin gene expression. There are two isoforms of C1orf77 that are produced as a result of alternative splicing events. The gene encoding C1orf77 maps to human chromosome 1, the largest human chromosome spanning about 260 million base pairs and making up 8% of the human genome. There are about 3,000 genes on chromosome 1, and considering the great number of genes there are also a large number of diseases associated with chromosome 1.

Small synthetic molecules called growth-hormone secretagogues (GHSs) stimulate the release of growth hormone (GH) from the pituitary through a G protein-coupled receptor (GHS-R). Recently, ghrelin ("ghre" is the Proto-Indo-European root of the word "grow") has been identified as an endogenous ligand for GHS-R. These peptides consist of 28 amino acids with an octanoyl-residue at Ser³, which is essential for expressing activity. Ghrelin stimulates GH release from rat primary culture pituitary cells in a dose-dependent manner and induces an increase in intracellular Ca²⁺ in GHS-R-expressing cells with an EC₅₀ value of 2.5 nM. Human ghrelin is homologous to rat ghrelin apart from two amino acids. The occurrence of ghrelin in both rat and human indicates that GH release from the pituitary may be regulated not only by hypothalamic GHRH, but also by ghrelin. In future, ghrelin might be used in the treatment of aging patients as well as in patients with idiopathic GH deficiency, obesity, osteoporosis or cardiovascular diseases.

Selumetinib (AZD6244) is a second-generation, orally available, potent and selective ATP non-competitive inhibitor of MEK 1/2. In direct assays, AZD6244 selectively inhibits purified active MEK1 and MEK2 with an IC(50) of 14nM, and AZD6244 has reported IC(50) values of <lt/>100nm for MEK1 in ELISA assays and in bindings assays against recombinant domains of MEK2, Kd values were 53nM for AZD6244.
