The BTB domain, also known as the POZ (poxvirus and zinc finger) domain, is an N-terminal homodimerization domain that contains multiple copies of kelch repeats and/or C2H2-type zinc fingers. Proteins that contain BTB domains are thought to be involved in transcriptional regulation via control of chromatin structure and function. RCBTB1 (regulator of chromosome condensation (RCC1) and BTB (POZ) domain containing protein 1), also known as GLP, CLLD7, CLLL7 or E4.5, is a 531 amino acid protein that localizes to the nucleus and contains two BTB (POZ) domains and six RCC1 repeats. Expressed ubiquitously, RCBTB1 is thought to be involved in cell cycle regulation, specifically via chromatin remodeling. The gene encoding RCBTB1 maps to a region on human chromosome 13 that is frequently deleted in B-cell chronic lymphocytic leukemia, suggesting a possible role for RCBTB1 in tumor suppression.

The nerve growth factor-induced clone C (NGFI-C/EGR4) gene is a zinc-finger transcription factor that is rapidly induced by nerve growth factor in rat pheochromocytoma PC12 cells and by seizure in brain. NGFI-C/EGR4 is closely related to the previously described early response genes, nerve growth factor-induced clone A (NGFI-A or EGR1), EGR2, and EGR3. These four early response (immediate early) proteins all contain very similar zinc-finger DNA binding domains and five highly homologous subdomains. EGR-4 functionally cooperate with NFAT proteins and induce expression of IL-2 and TNFalpha. Early growth response proteins (EGR) and nuclear factors of activated T cells (NFAT) form heterodimers and regulate proinflammatory cytokine gene expression.

Superoxide dismutase (SOD) is an antioxidant enzyme involved in the defense system against reactive oxygen species (ROS). SOD catalyzes the dismutation reaction of superoxide radical anion (O2-) to hydrogen peroxide, which is then catalyzed to innocuous O2 and H2O by glutathione peroxidase and catalase. Several classes of SOD have been identified. These include intracellular copper, zinc SOD (Cu, Zn-SOD/SOD-1), mitochondrial manganese SOD (Mn-SOD/SOD-2) and extracellular Cu, Zn-SOD (EC-SOD/SOD-3). SOD1 is found in all eukaryotic species as a homodimeric 32 kDa enzyme containing one each of Cu and Zn ion per subunit. The manganese containing 80 kDa tetrameric enzyme SOD2, is located in the mitochondrial matrix in close proximity to a primary endogenous source of superoxide, the mitochondrial respiratory chain. SOD3 is a heparin-binding multimer of disulfide-linked dimers, primarily expressed in human lungs, vessel walls and airways. SOD4 is a copper chaperone for superoxide dismutase (CCS), which specifically delivers Cu to copper/zinc superoxide dismutase. CCS may activate copper/zinc superoxide dismutase through direct insertion of the Cu cofactor.

The Zyxin family of proteins contains five members: Ajuba, Limd1, LPP, TRIP6 and Zyxin. Limd1 (LIM domain-containing protein 1) is a ubiquitously expressed tumor suppressor containing 3 LIM zinc-binding domains. LIM domains consist of a cysteine-rich consensus sequence containing two distinct zinc-binding subdomains, which mediate protein-protein interactions. Limd1 interacts with the proteins SQSTM1, Rb, p62 and TRAF6. Limd1 was first identified when the deletion of its gene was noted in some cervical cancers. Limd1 blocks <i>in vitro</i> and <i>in vivo</i> tumor growth and is down-regulated in lung cancer. Limd1 may regulate osteoclast development under stressful conditions via its interactions with TRAF6 and p62.

KLF4 is a member of the Kruppel-like factor (KLF) family of zinc finger transcription factors. Members of this family have in common 3 contiguous C2H2-type zinc fingers at the carboxyl terminus that comprise the DNA-binding domain. KLF4 is highly expressed in skin and gut epithelial tissues, but is also found in various other cells and tissues, including vascular endothelial cells, lymphocytes, lung, and testis. It is an important regulator of the cell cycle, transcription, and cell differentiation. Together with Sox2, Oct4, and cMyc, KLF4 can induce the reprogramming of primary human fibroblasts to a pluripotent state. KLF4 and other transcription factors can be introduced into cells by DNA transfection, viral infection, or microinjection. Protein transduction using TAT fusion proteins represents an alternative methodology for introducing transcription factors into primary as well as transformed cells. Recombinant human KLF4-TAT is a 51.7 kDa protein containing 483 amino acid residues, including 13- residue C-terminal TAT peptide.

AT-motif binding factor 1 (ATBF1) binds to the AT-rich core sequence element in the human a-fetoprotein enhancer (1). Alternative splicing generates the ATBF1-A and ATBF1-B (2,3). While ATBF1-A contains a 920-amino acid extension at the N-terminus, both ATBF1-A and ATBF1-B contain 4 DNA-binding homeobox domains (2,3). Additionally, ATBF1-A contains 23 zinc finger motifs while ATBF1-B contains 18 zinc finger motifs (1–3). The N-terminal extension unique to ATBF1-A has transcriptional repressor activity (4). In the small intestine, ATBF1-A inhibits expression of the brushborder enzyme aminopeptidase-N through direct binding to the AT motif element (5). Besides functioning in transcription regulation, ATBF1 also functions in ATPase activity (6). ATPase activity associated with ATBF1-A is DNA/RNA-dependent and requires both homeobox domains and zinc finger motifs (6). ATBF1 is highly expressed in spleen and brain tissues (7). The gene encoding human ATBF1 maps to chromosome 16q22.3-q23.1 (8).

This gene is closely linked to a gene encoding zinc finger protein of the cerebellum 5, a related family member on chromosome 13.
