Matrix metalloproteinases (MMP’s) belong to a family of secreted or membrane-associated zinc endopeptidases capable of digesting extracellular matrix components. MMP-1 (collagenase-1) is involved in tumor development and metastasis and rheumatoid arthritis. It is proposed as a therapeutic target for these diseases. Native pro-MMP-1 is prepared from culture medium of human rheumatoid synovial fibroblasts. MMP-1 is secreted as pro-enzyme, which consists of a propeptide of 80 amino acids, a catalytic domain of 162 amino acids, a 16-residue linker region, and a hemopexin domain of 189 amino acids. The native pro-MMP-1 has a major Mr 52-kDa unglycosylated and a minor Mr 57-kDa glycosylated form. The proteolytic activation of the 57/52-kDa species will form 47/42-kDa active collagenase, and a 22-kDa C-terminal fragment
The apparent Mr on SDS-PAGE is approximately 56kDa/52 kDa. The pro-MMP-1 can be fully activated by incubating with 1 mM APMA at 37°C for 3 hr. Its activity can be measured by FRET peptides. 10-20 ng of enzyme is sufficient for FRET-based assay

The Ovo family of zinc-finger transcription factors encode evolutionarily conserved genes including those from Caenorhabditis elegans, Drosophila melanogaster, mouse and human. Members of the Ovo family include Ovol1 and Ovol2. Ovol1 acts as a transcriptional repressor by interacting with key developmental signaling pathways such as Wnt and TGF-∫/BMP. Specifically, Ovol1 represses c-Myc and Id2 genes and establishes a balance between proliferation and differentiation of progenitor cells. Deletion of Ovol1 in mice leads to germ cell degeneration and defective sperm production in adult males. Ovol1 has also been shown to repress itself as well as Ovol2, which is thought to regulate neural development and vascular angiogenesis during embryogenesis.

CRP2 is a 193 amino acid nuclear protein that belongs to the CRP family of LIM domain proteins. Highly expressed in smooth muscle of aorta, CRP2 is thought to have a role in embryonic vascular system development and is downregulated following cell injury or PDGF-B exposure. CRP2 contains two LIM zinc-binding domains and is encoded by a gene that maps to human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome. Chromosome 12 is associated with a variety of diseases and afflictions, including hypochondrogenesis, achondrogenesis, Kniest dysplasia, Noonan syndrome and trisomy 12p, which causes facial developmental defects and seizure disorders.

This antibody recognizes a 66 kDa protein, which is identified as Tripartite motif-containing protein 29 (TRIM29). It interacts with the intermediate filament protein vimentin, a substrate for the PKC family of protein kinases, and with hPKCI-1, an inhibitor of the PKCs. TRIM29 protein contains both zinc finger and leucine zipper motifs, suggesting that the it may form homodimers and possibly associate with DNA. High expression of TRIM29 has been reported in gastric cancer and pancreatic cancer, and correlates with enhanced tumor growth and lymph node metastasis. TRIM29 is also able to distinguish lung squamous cell carcinoma from lung adenocarcinoma with ~90% positive accuracy, when used in a panel with TTF-1, p63, CK5/6, and Napsin-A antibodies.
