RNA polymerase II (Pol II) is an enzyme that is composed of twelve subunits and is responsible for the transcription of protein-coding genes. Transcription initiation requires Pol II-mediated recruitment of transcription machinery to a target promoter, thereby allowing transcription to begin. The largest subunit of Pol II (referred to as RPB1 or RPB205) is a 1,840 amino acid protein that contains one C2H2-type zinc finger and a C-terminal domain comprised of several heptapeptide repeats. Although Pol II function requires the cooperation of all twelve subunits, the largest subunit conveys Pol II catalytic activity and, together with the second largest subunit, forms the active center of the Pol II enzyme. Additionally, the large subunit participates in forming the DNA-binding domain of Pol II, a groove that is necessary for transcription of the DNA template. Without proper function of the large subunit, mRNA synthesis and subsequent transcription elongation cannot occur.

The Sp transcription factor family includes Sp1, Sp2, Sp3 (SPR-2) and Sp4 (SPR-1). Sp transcription factors share similar structures but do not share simi-lar functions. All four proteins contain a highly conserved DNA-binding domain composed of three zinc fingers at the C-terminus. Sp family members bind the consensus sequence GGGGCGGGGC and other closely related sequences which are known as GC boxes. Sp1, Sp3 and Sp4 share a high affinity for GC boxes while Sp2 does not. PLEKHM3 (pleckstrin homology domain containing, family M, member 3), also known as DAPR or PLEKHM1L (pleckstrin homology domain containing, family M, member 1-like), is a 761 amino acid phosphoprotein that contains two pleckstrin homology (PH) domains and one phorbol-ester/DAG-type zinc finger. Conserved in chimpanzee, dog, cow, mouse, rat, chicken and zebrafish, PLEKHM3 exists as three alternatively spliced isoforms that participate in metal ion binding. The gene that encodes PLEKHM3 maps to human chromosome 2q33.3. As the second largest human chromosome, chromosome 2 makes up approximately 8% of the human genome and contains 237 million bases encoding over 1,400 genes. Chromosome 2 contains a probable vestigial second centromere, as well as vestigial telomeres, which gives credence to the hypothesis that human chromosome 2 formed as a result of an ancient fusion of two ancestral chromosomes, which are still present in modern day apes.

Superoxide dismutase (SOD) is responsible for the elimination of cytotoxic active oxygen by catalyzing the dismutation of the superoxide radical to oxygen and hydrogen peroxide. There are three SOD isoenzymes in mammalian cells. They are: extracellular SOD (EC SOD), copper and zinc-containing SOD (Cu/Zn SOD) and manganese-containing SOD (Mn SOD). The Cu/Zn form contains Cu and Zn ions and exists as a 32 kDa dimer in the cytosol. Mn SOD is an 80 kDa tetramer that contains Mn ion and resides in the mitochondrial matrix. Mn SOD is a tumor necrosis factor (TNF)-inducible enzyme that protects cells from TNF-mediated apoptosis via superoxide anion detoxification and the subsequent regulation of apoptosis through cytochrome c release and the modulation of the redox state of the mitochondria. Mn SOD has also been shown to be a tumor suppressor in human breast cancer. Overexpression of this enzyme protects neurons from NMDA- and nitric oxide-induced neurotoxicity.
