Zinc finger FYVE domain-containing protein 27 (ZFYVE27), also known as SPG33, is a 411 amino acid member of the FYVE-finger family of proteins. The FYVE domain is a cysteine-rich domain of about 70 amino acids that plays a role in the endosomal localization of the FYVE-finger proteins, and a majority of these proteins serve as regulators of endocytic membrane trafficking. ZFYVE27, a multi-pass membrane protein, is an endosomal protein that binds to Spastin, a protein that is primarily involved in microtubule dynamics and severing, vesicular trafficking and endosomal trafficking. Mutations in the gene encoding ZFTVE27 affect neuronal intracellular trafficking in the corticospinal tract and are thought to lead to hereditary spastic paraplegia (HSP), a neurodegenerative disorder, characterized by progressive paralysis of the legs, which is caused by impaired axonal transport. Five isoforms of ZFYVE27 exist as a result of alternative splicing events.

Description:
TNFAIP3, also known as A20, is located in chromosome band 6q23, a region that is often deleted in B cell lymphomas. Recently, it was identified as a tumor suppressor gene in Hodgkin lymphoma and several subtypes of non-Hodgkin lymphomas. TNFAIP3 was initially identified as a zinc-finger protein that is rapidly and transiently induced by TNF-a, inhibiting NF-kB-dependent gene expression, and protecting cells from TNF-a-cytotoxicity. Overexpression of TNFAIP3 also inhibits the TLR2- and TLR4-mediated interleukin-8 synthesis in airway epithelial cells, suggesting that TNFAIP3 also acts as a negative regulator of TLR-mediated inflammatory responses, thereby protecting the host against harmful over-responses to pathogens. At least two isoforms of TNFAIP3 are known to exist.

ZNF318 is a 2279 amino acid endocrine regulatory protein that localizes to the nucleus. Highly expressed in testis, ovaries and kidneys, ZNF318 is a co-repressor of androgen receptor (AR)-mediated transcriptional activation and is thought to regulate transcription during spermatogenesis. ZNF318 interacts with the N-terminal domain of AR and contains two matrin-type zinc fingers. Two isoforms of ZNF318, designated TZF and TZF-L, are produced due to alternative splicing events. Each of these splice variants are thought to have unique roles in transcriptional regulation. While the TZF isoform functions as a repressor of AR-mediated transcriptional activation, the TZF-L isoform is thought to enhance AR-mediated transcriptional activation.

The Ovo family of zinc-finger transcription factors encode evolutionarily conserved genes including those from Caenorhabditis elegans, Drosophila melanogaster, mouse and human. Members of the Ovo family include Ovol1 and Ovol2. Ovol1 acts as a transcriptional repressor by interacting with key developmental signaling pathways such as Wnt and TGF-∫/BMP. Specifically, Ovol1 represses c-Myc and Id2 genes and establishes a balance between proliferation and differentiation of progenitor cells. Deletion of Ovol1 in mice leads to germ cell degeneration and defective sperm production in adult males. Ovol1 has also been shown to repress itself as well as Ovol2, which is thought to regulate neural development and vascular angiogenesis during embryogenesis.

Highly expressed in smooth muscle of aorta, CRP2 is thought to have a role in embryonic vascular system development and is downregulated following cell injury or PDGF-B exposure. CRP2 contains two LIM zinc-binding domains and is encoded by a gene that maps to human chromosome 12, which encodes over 1,100 genes and comprises approximately 4.5% of the human genome. Chromosome 12 is associated with a variety of diseases and afflictions, including hypochondrogenesis, achondrogenesis, Kniest dysplasia, Noonan syndrome and trisomy 12p, which causes facial developmental defects and seizure disorders.
