The GTPase of the immunity-associated protein (GIMAP) family of proteins include seven members that are expressed by genes residing on human chromosome 7. GIMAP proteins have been implicated in the regulation of lymphomyeloid cell survival. GIMAP1 (GTPase, IMAP family member 1), also known as IMAP1 (immunity-associated protein 1), HIMAP1 or IMAP38, is a 306 amino acid single-pass type IV membrane protein of the endoplasmic reticulum (ER) belonging to the GIMAP family. Encoded by a gene located on human chromosome 7, GIMAP1 is expressed primarily in spleen with lower levels found in lymph node. Chromosome 7 houses over 1,000 genes, comprises nearly 5% of the human genome and has been linked to Osteogenesis imperfecta, Pendred syndrome, Lissencephaly, Citrullinemia and Shwachman-Diamond syndrome.

Defects in G6PD are the cause of chronic non-spherocytic hemolytic anemia (CNSHA) . Deficiency of G6PD is associated with hemolytic anemia in two different situations. First, in areas in which malaria has been endemic, G6PD-deficiency alleles have reached high frequencies (1% to 50%) and deficient individuals, though essentially asymptomatic in the steady state, have a high risk of acute hemolytic attacks. Secondly, sporadic cases of G6PD deficiency occur at a very low frequencies, and they usually present a more severe phenotype. Several types of CNSHA are recognized. Class-I variants are associated with severe NSHA; class-II have an activity <10% of normal; class-III have an activity of 10% to 60% of normal; class-IV have near normal activity.

The apolipoprotein L gene family maps to a region on chromosome 22 and encodes six highly homologous proteins designated apoL-I, apoL-II, apoL-III, apoL-IV, apoL-V and apoL-VI, all of which function as components of plasma lipoproteins. ApoL-V (apolipoprotein L-V), also known as APOL5, is a 433 amino acid protein that localizes to the cytoplasm and belongs to the apolipoprotein L family. Expressed in a variety of tissues including testis, stomach, uterus and skeletal muscle, apoL-V is thought to affect the movement of lipids in the cytoplasm and may allow the binding of lipids to organelles. Like other members of the apolipoprotein L family, apoL-V is thought to be involved in the development of schizophrenia.

In muscle tissue, collagen serves as a major component of the endomysium. Collagen constitutes one to two percent of muscle tissue, and accounts for 6% of the weight of strong, tendinous muscles. A collagen may be defined as a protein containing sizable domain(s) of triple-helical conformation. Type IV collagen is a major macromolecular constituent of basement membranes and can be readily isolated from basement-membrane-rich tissues or highly vascularized tissues such as the placental villi. This collagen appears to be largely restricted to structures identifiable as basement membranes. In contrast, type VI collagen appears to be prevalent in several tissues even though it has been isolated largely from placental villi preparations. The extent to which type VII and VIII collagens are distributed is not known.

GLI2 encodes a protein which belongs to the C2H2-type zinc finger protein subclass of the Gli family. Members of this subclass are characterized as transcription factors which bind DNA through zinc finger motifs. These motifs contain conserved H-C links. Gli family zinc finger proteins are mediators of Sonic hedgehog (Shh) signaling and they are implicated as potent oncogenes in the embryonal carcinoma cell. The protein encoded by this gene localizes to the cytoplasm and activates patched Drosophila homolog (PTCH) gene expression. It is also thought to play a role during embryogenesis. The encoded protein is associated with several phenotypes- Greig cephalopolysyndactyly syndrome, Pallister-Hall syndrome, preaxial polydactyly type IV, postaxial polydactyly types A1 and B.

The Semphorins are a large family of phylogenetically conserved proteins that play a pivotal role in maintaining homeostasis in the immune system. Twenty members of this family have been identified and categorized into eight subclasses based on sequence similarity and distinctive structural features. CD100, also known as Sema4D, is a 150 kDa transmembrane class IV semaphorin. Studies have shown that CD100 can induce monocyte migration, T-cell activation, and B-cell survival, as well as T/B cell and T/DC "cooperation”. The CD100 precursor contains 862 amino acids, including a 21 a.a. signal sequence, a 713 a.a. extracellular domain, a 21 a.a transmembrane sequence, and a 107 a.a cytoplasmic region. The extracellular sequence contains several structural features, including a 479 a.a "sema” domain, a 79 a.a. Ig-like sequence, and a 52 a.a "Plexin-type repeat”. Recombinant soluble CD100 is a 78.9 kDa protein comprising the extracellular domain of CD100 (711 amino acids). SDS-PAGE analysis run under non-reducing conditions shows a mixture of disulfide linked dimer and monomer.
