Spondin-2, also referred to as mindin, belongs to the F-spondin family of secreted extracellular matrix proteins. Spondins are characterised by the presence of F-spondin domains 1 and 2 (FS1 and FS2) at the N-terminus and a thrombospondin-type 1 repeat (TSR1) domain at the C-terminus. Spondin-2 functions as a pattern-recognition molecule for bacterial and viral pathogens and as an integrin ligand for inflammatory cell recruitment and T cell priming. In addition to its roles in promoting neuron outgrowth and inhibiting both cancer and angiogenesis, Spondin-2 plays an important role in the initiation of the immune response and is involved in inflammatory processes.

The prolactin receptor (PRLR) is a member of the class I cytokine/lactogen receptor family which mediates the diverse cellular actions of prolactin in several tissues. PRLRs are expressed in normal and neoplastic human breast tissue, and in most breast cancer cells. PRLR contains an extracellular region that binds prolactin, a transmembrane region, and a cytoplasmatic region required for the activation of the Jak2–Stat5 signal transduction pathway by Prl which is essential for transcriptional activation of all known prolactin regulated genes. PRLRs have also been observed in ovarian follicular cells of mice, pigs, sheep, deer, and humans, as well as in luteal tissue in cow and horse ovaries. Furthermore, PRLR knockout mice exhibit failure of embryonic implantation, reduced number of mature oocytes, and low fertilization rates. Knockout females also display a reduced number of primary follicles.

OX40L is expressed by DCs, macrophages and B cells and signals via its cognate receptor OX40 which is mainly expressed on APCs. OX40L/OX40 interactions are important in T-cell activation and survival and for the generation of memory T cells from activated effector T cells. OX40L–OX40 co-stimulation leads to activation of TNF receptor associated factor (TRAF) 2, 3 and 5. This pathway has been shown to prolong the survival of effector CD4+Th cells as well as contributes to generation of memory T cells.

TREM-2 is one member of the TREM family, inhibiting the releasing of inflammatory mediators, so it is an important in vivo anti-inflammatory receptor. TREM-2 consists of an 18 aa signal sequence, a 153 aa extracellular domain (ECD) with one V-type Ig-like domain, a 21 aa transmembrane (TM) domain, and a 35 aa cytoplasmic tail. A soluble form of TREM-2 (TREM-2b) created by alternate splicing diverges at aa 161. TREM-2 transduces intracellular signals through the adaptor DAP12. After binding of TREM-2 with ligand, the TREM-2/DAP12 (dead-cell-activated-receptor-associated protein)-mediated signal transduction pathway causes a series of intracellular protein tyrosine phosphorylation reactions and enzymatic reactions, which then activate the myeloid cells and participate T cell responses.

Cochlin is an extracellular matrix protein expressed in the cochlea and vestibule of the inner ear. Mutations in the cochlin gene are linked to the autosomal-dominant DFNA9 syndrome, a disorder characterized by hearing loss. Cochlin is also expressed in follicular dendritic cells of the spleen and the lymph nodes. The N-terminus of cochlin contains a LCCL domain also present in Factor C, a serine protease of the horseshoe crab Limulus, involved in activating the coagulation cascade in response to LPS. A recent study showed that in response to inflammation, LPS or bacterial infection, cochlin is cleaved by the proteases aggrecanases and secreted into blood circulation. The secreted cochlin (LCCL domain) seems to have a role in local cytokine production, recruitment of immune effector cells and bacterial clearance. Cochlin is secreted as two forms p8 and p18, whereas p18 being the glycosylated form of p8.

CD28 and CTLA-4 together with their ligands, CD80 (B7-1) and CD86 (B7-2), constitute one of the dominant costimulatory pathways that regulate T and B cell responses. CD28 and CTLA-4 are structurally homologous molecules that are members of the immunoglobulin (Ig) gene superfamily. Both CD28 and CTLA-4 are composed of a single Ig V-like extracellular domain, a transmembrane domain and an intracellular domain. CD28 and CTLA-4 are both expressed on the cell surface as disulfide-linked homodimers or as monomers. The genes encoding these two molecules are closely linked on human chromosome 2 and mouse chromosome 1. Mouse CD28 is expressed constitutively on virtually 100% of mouse T cells and on developing thymocytes. Cell surface expression of mouse CD28 is downregulated upon ligation of CD28 in the presence of PMA or PHA. In contrast, CTLA-4 is not expressed constitutively but is upregulated rapidly following T cell activation and CD28 ligation. Cell surface expression of mouse CTLA-4 peaks approx.y 48 hours after activation. Although both CTLA-4 and CD28 can bind to the same ligands, CTLA-4 binds to B7-1 and B7-2 with a 20-100 fold higher affinity than CD28. CD28/B7 interaction has been shown to prevent apoptosis of activated T cells via the upregulation of bcl-XL. CD28 ligation has also been shown to regulate Th1/Th2 differentiation.
