The Hoechst stains are a family of fluorescent stains for labeling DNA in fluorescence microscopy. The blue fluorescent Hoechst dye is a cell permeable nucleic acid stain that has multiple applications, including sensitive detection of DNA in the presence of RNA in agarose gels, automated DNA determination, sensitive determination of cell number and chromosome sorting. Useful vital stain for the flow cytometric recognition of DNA damage and other viability measurements by monitoring the emission spectral shifts of the dyes. Because this fluorescent stain labels DNA, it can also be used to visualize nuclei and mitochondria. Hoechst 34580 can be excited by violet laser at 405 nm, and emit blue/cyan fluorescence light around an emission maximum at ~440 nm. It is cell-permeable and an excellent tool for flow cytometry studies.

The conversion from tyrosine to 4-HPPA is catalyzed by tyrosine aminotransferase. 4-HPPA can be converted to homogentisic acid which is one of the precursors to ochronotic pigment. A deficiency in the catalytic activity of HPD is known to lead to tyrosinemia type III, an autosomal recessive disorder characterized by elevated levels of blood tyrosine and massive excretion of tyrosine derivatives into urine.

Water soluble pH indicator used in the 6.8 (yellow) to 8.2 (red) range and frequently used in cell biology laboratories. Widely used as cell tissue culture pH marker. A small amount of phenol red added to the growth medium will have a pink-red color under normal conditions. Typically, 15 mg/l are used for cell culture. In the event of problems, waste products produced by dying cells or overgrowth of contaminants will cause a change in pH, leading to a change in indicator color. For example, a culture of relatively slowly dividing mammalian cells can be quickly overgrown by bacterial contamination. This usually results in an acidification of the medium, turning it yellow. The waste products produced by the mammalian cells themselves will slowly decrease the pH, gradually turning the solution orange and then yellow. This color change is an indication that even in the absence of contamination, the medium needs to be replaced. Shown to be a weak estrogen mimic (possible through inpurities), which can enhance the growth of cells that express the estrogen receptor in cell cultures. Might be useful as a differentiation factor.

RIG-I (retinoic acid-inducible gene I; Ddx58) and MDA5 (melanoma differentiation-associated gene 5, also known as Ifih1 or Helicard) are proteins that sense viral replication intermediates, such as double-stranded RNA and triggers the host antiviral programs. These molecules signal the downstream activation of NF-kappaB and IFN regulatory factor (IRF) -3, which coordinately regulate the expression of type-I interferons. Cardif (also called VISA/IPS-1/MAVS) is a new CARD (caspase activation and recruitment domain)-containing adaptor protein that interacts with the CARD domain of RIG-I and MDA5, leading to the activation of NF-kappaB and IRF3. Cardif is located to the mitochondrial outer membrane. Removal of the mitochondrial-targeting domain of cardif abolishes its ability to induce IFNs. Cardif is cleaved and inactivated by NS3-4A, a serine protease from hepatitis C virus known to block interferon-beta production.
