Description:
IL-16 is a CD8+ T cell-derived cytokine that induces chemotaxis of CD4+ T cells, CD4+ monocytes, and eosinophils. Analysis by gel filtration suggests that, under physiological conditions, hIL-16 exists predominantly as a noncovalently linked multimer, but that some IL-16 may exist as a monomer. However, only the multimeric form appears to possess chemotactic activity, suggesting that receptor cross-linking may be required for activity. IL-16 also induces expression of IL-2 receptor (IL-2R) and MHC class II molecules on CD4 + T cells. Human and murine IL-16 show significant cross-species reactivity. Recombinant Human IL-16 is a 12.4 kDa protein consisting of 121 amino acid residues.

CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. It is transiently expressed at a low level during B cell development in bone marrow on the CD45R/B220low TdT- sIg- pre-B II and CD45R/B220low TdT- sIgM+sIgD- immature B cells, but not on the CD45R/B220low TdT+ sIg- pro-B/pre-B I stage nor on CD45R/B220high TdT- sIgM+sIgD+ mature B cells. CD25 is expressed at a higher level during very early T-cell development in fetal and adult thymus. The 7D4 monoclonal antibody reacts with an epitope between amino acids 125 and 212 that is distinct from that recognized by the monoclonal antibody 3C7 (SB Cat. No. 1600). While the monoclonal antibody 7D4 does not block the binding of IL-2 to CD25, when used in combination with the monoclonal antibody 3C7 it results in higher levels of inhibition of IL-2-driven proliferation than either of these two monoclonal antibodies alone.

Interleukin 15 (IL-15RA) is a cytokine receptor that specifically binds IL-15 with high affinity. IL-15 regulates T and natural killer cell activation and proliferation. The IL-15 and IL-2 receptors share two subunits, IL-2R beta and IL-2R gamma, and IL-15RA is structurally related to IL-2RA, an additional IL-2-specific alpha subunit necessary for high affinity IL-2 binding. Unlike IL-2RA, IL-15RA is capable of binding IL-15 with high affinity independent of the other subunits. IL-15RA is thought to enhance cell proliferation and expression of apoptosis inhibitor Bcl-xL and Bcl-2.
