Ca2+-indicator fluo-3 for use with visible-light excitation sources in flow cytometry and confocal laser-scanning microscopy. Since about 1996, fluo-3 has also been extensively used in cell-based high-throughput screening assays for drug discovery. Fluo-3 AM is essentially nonfluorescent until it is hydrolyzed intracellularly and/or in the presence of Ca2+. The green-fluorescent emission (~525 nm) of Ca2+-bound fluo-3 is conventionally detected using optical filter sets designed for fluorescein (FITC). As prepared by Minta, Kao and Tsien, fluo-3 was originally reported to undergo an ~40-fold increase in fluorescence upon binding Ca2+.

The claudin superfamily consists of many structurally related proteins that are important structural and functional components of tight junctions in paracellular transport. Claudins are located in both epithelial and endothelial cells in all tight junction-bearing tissues. Claudins, which consist of four transmembrane domains and two extracellular loops, make up tight junction strands. It is suggested that the claudin family of proteins regulate transport through tight junctions via differential discrimination for solute size and charge. Claudin expression is often highly restricted to specific regions of different tissues and may have an important role in transcellular transport through tight junctions. Claudin-22, also known as CLDN22, is a 220 amino acid member of the claudin family that participates in tight junction-specific obliteration of the intercellular space through calcium-independent cell-adhesion activity.

Leptin receptor (OB-R) is a type I cytokine receptor family protein with significant amino acid sequence identity with gp130, G-CSF receptor, and LIF receptor. An OB-R transcript encodes a soluble form of the receptor. The transcript was reported to be expressed predominantly in regions of the hypothalamus previously thought to be important in body weight regulation. OB-R binds leptin with high affinity and is a potent leptin antagonist. Human OB-R encodes a 1 165 amino acid residue precursor protein with a 22 amino acid residue signal peptide, an 816 amino acid residue extracellular domain, a 21 amino acid residue transmembrane domain and a 303 amino acid residue cytoplasmic domain.
