Description:
DNA methylation in vertebrate animals is an epigenetic modification that is important for embryonic development, imprinting, and the inactivation of X chromosomes. DNA methylation is catalyzed by a family of DNA methyltransferases (DNMTs) that include the maintenance enzyme DNMT1 and de novo methyltransferases DNMT3a and DNMT3b. The overexpression of DNMT1, DNMT3a, and DNMT3b has been reported in various malignancies, including gastric, urothelial, and lung cancers, and may be related to tumorigenesis, tumor progression, and poor survival. Two isoforms of DNMT3a exist: the full-length DNMT3a, and the shorter form DNMT3a2 which lacks the N-terminal fragment. DNMT3a is expressed ubiquitously at low levels, while DNMT3a2 is specially expressed at high levels in embryonic stem cells and shows restricted expression in tissues known to undergo de novo methylation including testis and ovary. This antibody was raised against the N-terminal region of human DNMT3a. It is expected to detect the 120-130 kDa DNMT3a but not 72-100 kDa DNMT3a2.

It specifically binds to the histone H3 N-terminal tail when methylated on K9 by two functional domains, a PHD finger that defines the binding specificity and an SRA (SET- and RING-associated) domain that promotes binding activity. UHRF1BP1 is required for proper heterochromatin formation in mammalian cells. Furthermore, UHRF1BP1 is thought to be a pivotal target for the ERK1/2 signaling pathway to control the proliferation of Jurkat T cells.

DPF3 has been shown to be a epigenetic key factor for heart and muscle development and can bind to methylated and acetylated lysine residues of histone 3 and 4, suggesting that DPF3 may play a role in recruiting chromatin remodeling complexes to acetylated histones. Dact3 Antibody: The Wnt signaling cascade is a conserved process in multicellular animals that plays important roles during development and can contribute to cancer and other diseases. Many members of this pathway are also expressed in the postnatal tissues such as brain. One such protein is Dact3, a member of the Dact protein family that was initially identified through binding to Disheveled (Dvl), a cytoplasmic protein essential to Wnt signaling. Dact3 is expressed in the ventral region of maturing somites, limb bud and branchial arch mesenchyme, embryonic CNS, and the adult brain. Recent evidence shows that Dact3 acts as a negative regulator Wnt/beta-catenin signaling that is repressed at the transcriptional level in colorectal cancer and this repression is associated with bivalent histone modifications. This repression can be reversed by pharmacological agents that targets both histone methylation and deacetylation, suggesting that Dact3 may be a potential target for therapeutic treatment of this cancer. At least three isoforms of Dact3 are known to exist.
