KEAP1 Antibody: KEAP1 (kelch-like ECH-associated protein 1) is a stress sensing adaptor for the Cullin3 (Cul3)-dependent E3 ubiquitin ligase complex that negatively regulates NRF2 (NF-E2-related factor 2) and plays a role in the oxidative stress response. It targets NFE2L2/NRF2 for ubiquitination and degradation by the proteasome. KEAP1 contains an amino terminal BTB/POZ domain and a carboxyl terminal KELCH domain which are required for interaction with NRF2, and in binding Cul3-E3 ubiquitin ligase. Altered expression of NRF2 is associated with chronic obstructive pulmonary disease (COPD). KEAP1 also targets the down regulation of NF-kappa B activity by targeting IKKbeta ; degradation. Mutation of the KEAP1 gene is found in lung cancer.

PDIA1 (protein disulfide isomerase family A member 1) is the beta subunit of prolyl 4-hydroxylase, a highly abundant multifunctional enzyme that belongs to the protein disulfide isomerase family. When present as a tetramer consisting of two alpha and two beta subunits, this enzyme is involved in hydroxylation of prolyl residues in preprocollagen. PDIA1 is also a disulfide isomerase containing two thioredoxin domains that catalyze the formation, breakage and rearrangement of disulfide bonds. Other known functions include its ability to act as a chaperone that inhibits aggregation of misfolded proteins in a concentration-dependent manner, its ability to bind thyroid hormone, its role in both the influx and efflux of S-nitrosothiol-bound nitric oxide, and its function as a subunit of the microsomal triglyceride transfer protein complex.

CLEC7A, also known as dectin-1, is a member of the C-type lectin/C-type lectin-like domain (CTL/CTLD) superfamily and is predominantly expressed on myeloid cells. It is a small glycoprotein type II membrane receptor with an extracellular C-type lectin-like domain fold and a cytoplasmic domain with an immunoreceptor tyrosine-based activation motif (ITAM). CLEC7A functions as a pattern-recognition receptor that recognizes a variety of beta-1,3-linked and beta-1,6-linked glucans from fungi and plants, and in this way plays a role in innate immune response. Upon fungal exposure, CLEC7A activates Syk tyrosine kinase, triggering a massive oxidative burst through the formation of reactive oxygen species.

ADH6 (alcohol dehydrogenase 6), also known as ADH-5, is a 368 amino acid member of the class V zinc-containing alcohol dehydrogenase family. This family of enzymes functions to metabolize a wide variety of substrates such as retinol, hydroxysteroids, ethanol, aliphatic alcohols and lipid peroxidation products. Localized to the cytoplasm and expressed in the stomach and liver, ADH6 catalyzes the reversible oxidation of alcohols to their corresponding aldehydes or ketones and is able to bind two zinc ions as cofactors. ADH6 contains a glucocorticoid response element upstream of its 5' UTR which is thought to be a steroid binding site, suggesting that expression of ADH6 may be under hormonal control. Multiple isoforms of ADH6 exist due to alternative splicing events.

IL-23 Antibody: Like interleukin-27 (IL-27), IL-23 is a recently discovered member of the IL-6/IL-12 family of proinflammatory and immunoregulatory cytokines. It exists as a heterodimer composed of the IL-12p40 subunit and a novel p19 subunit. IL-23 is secreted by activated dendritic cells, macrophages, and monocytes. Its biological activities include enhancing the proliferation of memory T cells and the production of IFN-gamma, IL-12, and TNF-alpha from activated T cells, and can stimulate macrophages to produce TNF-alpha and nitric oxide. It has also been shown to possess potent anti-tumor and anti-metastatic activity in mouse models of cancer, suggesting a potential role for IL-23 in therapeutic treatment of cancer.

FAM120A (C9orf10) is a member of the constitutive coactivator of PPAR gamma family and the gene was mapped to chromosome 9q22.31. FAM120A was recently detected within the Pur-alpha-containing mRNA-protein complex in the brain. As a novel RNA-binding protein, FAM120A is a critical component of the oxidative stress-induced survival signaling. It may participate in mRNA transport in the cytoplasm. FAM120A activates src family kinases and acts as a scaffolding protein enabling src family kinases to phosphorylate and activate PI3-kinase. FAM120A protects cells from apoptosis through activation of SRKs in response to oxidative stress. Blocking of the survival signaling mediated by FAM120A, which sensitizes the cancer cells to stress-induced apoptosis, may be a novel therapeutic approach for gastric scirrhous carcinoma cells.

It also functions as a scavenger receptor, mediating macrophage uptake of oxidized low-density lipoprotein (LDL) and recognition of apoptotic polymorphonuclear leukocytes (PMN).
