Description:
GATA1 is a Cys2/Cys2 zinc finger DNA binding protein that is expressed primarily in erythroid, megakaryocytic, mast cells and eosinophilic cells. It belongs to the GATA family of transcription factors. GATA1 is a transcriptional activator which probably serves as a general switch factor for erythroid development. It binds to DNA sites with the consensus sequence [AT]GATA[AG] within regulatory regions of globin genes and of other genes expressed in erythroid cells. The protein also plays an important role in erythroid development by regulating the switch from fetal hemoglobin production to adult hemoglobin.

Description:
ELAC2 is a zinc phosphodiesterase which displays some tRNA 3' processing endonuclease activity. It is thought to be involved in tRNA maturation, acting by removing a 3' trailer from precursor tRNA. ELAC2 also interacts with activated Smad family member 2 (Smad2) and its nuclear partner forkhead box H1 (also known as FAST-1), and reduced expression can suppress transforming growth factor-beta induced growth arrest. Mutations in this gene result in an increased risk of prostate cancer. A number of allelic variants have been investigated in hereditary prostate cancer.

Description:
The protein encoded by this gene is a type I transmembrane protein that localizes to the endocytic pathway. This protein contains a RING zinc-finger motif and has been shown to possess E3 ubiquitin ligase activity. Expression of this gene in retrovirally transduced T cell hybridoma significantly inhibits activation-induced IL2 and IL4 cytokine production. Induced expression of this gene was observed in anergic CD4(+) T cells, which suggested a role in the induction of anergic phenotype. Alternatively spliced transcript variants encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008].

Description:
Hereditary spastic paraplegias (HSPs) are genetically and phenotypically heterogeneous disorders. Spastic paraplegia with thinning of the corpus callosum (ARHSP-TCC) is a relatively frequent form of complicated hereditary spastic paraplegia in which mental retardation and muscle stiffness at onset are followed by slowly progressive paraparesis and cognitive deterioration. SPG15 is the second gene known to be responsible for ARHSP-TCC in the Italian population. Mutations in this gene are associated with autosomal recessive spastic paraplegia-15. SPG15 encodes a protein containing a FYVE zinc finger binding domain which is thought to target these proteins to membrane lipids through interaction with phospholipids in the membrane. SPG15 mRNA is widely distributed in human tissues, as well as in rat embryos, suggesting a possible role for this protein during embryonic development. SPG15 co-localizes partially with endoplasmic reticulum and endosome markers, suggesting a role in intracellular trafficking. Multiple isoforms of SPG15 are known to exist.

Description:
Matrix metalloproteinases (MMPs) are a family of endoproteases that require zinc and calcium for expressing catalytic activity. These enzymes play a central role in the maintenance and remodeling of the extracellular matrix. Elevated expression of their activity, caused either by up-regulation of their expression or down-regulation of their cognate inhibitors, has been implicated in various degenerative disorders, including arthritis, cardiovascular disease, skeletal growth-plate disorders, and cancer metastasis. MMP-3 degrades fibronectin, laminin, collagens III, IV, and X, and cartilage proteoglycans. Recombinant Human MMP-3 is a 42.8 kDa protein containing the entire catalytic N-terminal domain and the C-terminal domain (378 amino acids).

Description:
Angiotensin-Converting Enzyme 2 (ACE-2) is an integral membrane protein and a zinc metalloprotease of the ACE family, the ACE family includes somatic and germinal ACE. ACE-2 cleaves angiotensins I and II as a carboxypeptidase, ACE-2 converts angiotensin I to angiotensin 1-9, and angiotensin II to angiotensin 1-7. ACE-2 is also able to hydrolyze apelin-13 and dynorphin-13 with high efficiency. ACE-2 can be high expressed in testis, kidney and heart, in colon, small intestine and ovary at moderate levels. Captopril and lisinopril as the classical ACE inhibitor don’t inhibit ACE-2 activity. ACE-2 may play an important role in regulating the heart function.
