Description:
The Extracellular regulated kinases, Erk-1 and Erk-2, both play a critical role in cellular proliferation. These extracellular kinases, are known to reside downstream of the Mitogen activated protein kinase/Extracellular protein kinases referred to as MEK. Both MEK-1 and MEK-2 phosphorylate and activate Erk on the conserved TEY motif within the activation loop of the kinase.

Description:
Bifunctional enzyme. The C-terminal domain has epoxide hydrolase activity and acts on epoxides (alkene oxides, oxiranes) and arene oxides. Plays a role in xenobiotic metabolism by degrading potentially toxic epoxides. Also determines steady-state levels of physiological mediators. The N-terminal domain has lipid phosphatase activity, with the highest activity towards threo-9,10-phosphonooxy-hydroxy-octadecanoic acid, followed by erythro-9,10-phosphonooxy-hydroxy-octadecanoic acid, 12-phosphonooxy-octadec-9Z-enoic acid, 12-phosphonooxy-octadec-9E-enoic acid, and p-nitrophenyl phospate.

Description:
Intercellular signal essential for a variety of patterning events during development: signal produced by the notochord that induces ventral cell fate in the neural tube and somites, and the polarizing signal for patterning of the anterior-posterior axis of the developing limb bud. Displays both floor plate- and motor neuron-inducing activity. The threshold concentration of N-product required for motor neuron induction is 5-fold lower than that required for floor plate induction. Activates the transcription of target genes by interacting with its receptor PTCH1 to prevent normal inhibition by PTCH1 on the constitutive signaling activity of SMO (By similarity).

Description:
S100A8 is a calcium- and zinc-binding protein which plays a prominent role in the regulation of inflammatory processes and immune response. It can induce neutrophil chemotaxis and adhesion. Predominantly found as calprotectin (S100A8/A9) which has a wide plethora of intra- and extracellular functions. The intracellular functions include: facilitating leukocyte arachidonic acid trafficking and metabolism, modulation of the tubulin-dependent cytoskeleton during migration of phagocytes and activation of the neutrophilic NADPH-oxidase. Activates NADPH-oxidase by facilitating the enzyme complex assembly at the cell membrane, transferring arachidonic acid, an essential cofactor, to the enzyme complex and S100A8 contributes to the enzyme assembly by directly binding to NCF2/P67PHOX. The extracellular functions involve proinfammatory, antimicrobial, oxidant-scavenging and apoptosis-inducing activities. Its proinflammatory activity includes recruitment of leukocytes, promotion of cytokine and chemokine production, and regulation of leukocyte adhesion and migration. Acts as an alarmin or a danger associated molecular pattern (DAMP) molecule and stimulates innate immune cells via binding to pattern recognition receptors such as Toll-like receptor 4 (TLR4) and receptor for advanced glycation endproducts (AGER). Binding to TLR4 and AGER activates the MAP-kinase and NF-kappa-B signaling pathways resulting in the amplification of the proinflammatory cascade. Has antimicrobial activity towards bacteria and fungi and exerts its antimicrobial activity probably via chelation of Zn(2+) which is essential for microbial growth. Can induce cell death via autophagy and apoptosis and this occurs through the cross-talk of mitochondria and lysosomes via reactive oxygen species (ROS) and the process involves BNIP3. Can regulate neutrophil number and apoptosis by an anti-apoptotic effect; regulates cell survival via ITGAM/ITGB and TLR4 and a signaling mechanism involving MEK-ERK.

Description:
Receptor for gastrin and cholecystokinin. The CKK-B receptors occur throughout the central nervous system where they modulate anxiety, analgesia, arousal, and neuroleptic activity. This receptor mediates its action by association with G proteins that activate a phosphatidylinositol-calcium second messenger system. Isoform 2 is constitutively activated and may regulate cancer cell proliferation via a gastrin-independent mechanism.

Interleukin 31 (IL-31) is an immunoregulatory cytokine that is expressed by activated type 2 T helper (Th2) cells. IL-31 signals through a heterodimer receptor consisting of the IL-31 Receptor A (IL-31RA) and the oncostatin M receptor (OSMR), which are expressed on monocytes, epithelial cells, and keratinocytes. IL-31 promotes allergic reactions and inflammatory skin diseases.

CD63 is expressed on activated platelets. CD63 is a lysosomal membrane glycoprotein that is translocated to plasma membrane after platelet activation. It is also present in monocytes and macrophages and is weakly expressed on granulocytes, B, and T cells. CD63 is identical to the melanoma-associated antigen which isME491 and to the platelet antigen PTLGP40.

Description:
May be the important intermediate by which p53/TP53 mediates its role as an inhibitor of cellular proliferation in response to DNA damage. Binds to and inhibits cyclin-dependent kinase activity, preventing phosphorylation of critical cyclin-dependent kinase substrates and blocking cell cycle progression. Functions in the nuclear localization and assembly of cyclin D-CDK4 complex and promotes its kinase activity towards RB1. At higher stoichiometric ratios, inhibits the kinase activity of the cyclin D-CDK4 complex.

Description:
RIG-I and MDA5 are highly conserved helicases involved in the innate immune response to virus. RIG-I is a member of the DEAD-box RNA helicases and is activated by cytoplasmic dsRNA and 5’-ppp RNA produced during the viral replication. The protein is characterized by a N-terminal region with two caspase recruitment domains (CARD) and a C-terminal region harboring potential ATP-dependent RNA helicase activity. RIG-I recruits the CARD adaptor inducing IFN-beta (Cardif) in a CARD-CARD-dependent manner resulting in NF-kappaB and IRF3 activation.

Description:
Mitogen activated protein kinase kinase 6 (MEK6 or MKK6) belongs to the serine/threonine protein kinase family and the MAPK kinase subfamily (MAP2K, MKK or MEKs). MEK6, closely related to MEK3, catalyzes the concomitant phosphorylation of a threonine and a tyrosine residue in MAP kinase p38, thus activating it, in response to inflammatory cytokines and environmental stress. As an essential component of p38 MAP kinase mediated signal transduction pathway, this protein is involved in many cellular processes such as stress induced cell cycle arrest, transcription activation and apoptosis.
