Description:
Nuclear factor kappa-B (NFk) B) functions as a sequence specific transcriptional activator that binds to the intronic enhancer of kappa light chain gene in B lymphocytes. NF-kB is a heterodimer consisting of a 50 kDa DNA binding subunit (p50) and a 65 kDa transactivation subunit (p65/RelA) (ref. 3), both of which exhibit sequence homology to the protooncogene c-Rel. p50 has an isoform called p49/p52, and both proteins derive from the amino-terminal of precursor protein p105 and p100, respectively. The p50/p65 heterodimer remains in inactive form in the cytosol as a complex with its inhibitor, IkB. Upon stimulation of cells by a variety of stimuli, including lipopolysaccharide (LPS), pro-inflammatory cytokines like interleukin-1 and tumor necrosis factor, and viral infection, IkB becomes phosphorylated and degraded by the proteosome. The active NF-kB heterodimer translocates into the nucleus and induces gene expression.

Plasminogen Activator Inhibitor-1 (PAI-1, Serpin E1) is a member of the serpin family of serine protease inhibitors, and is the primary inhibitor of urokinase and tissue plasminogen activator (tPA). PAI-1 is expressed predominantly in adipose, liver and vascular tissues, and is also produced by certain tumor cells. Elevated levels of PAI-1 are associated with obesity, diabetes and cardiovascular disease, and increased production of PAI-1 is induced by various obesity related factors such as TNFα, glucose, insulin, and very-low-density lipoprotein. The obesity related elevation of PAI-1 levels along with the consequential deficiency in plasminogen activators can lead directly to increased risk of thrombosis and other coronary diseases. Accordingly, PAI-1 has been implicated as an important molecular link between obesity and coronary disease. PAI-1 can also specifically bind vitronectin (VTN) to form a stable active complex with an increased circulatory half life relative to free PAI-1. Recombinant human PAI-1 is a 42.7 kDa protein containing 379 amino acid residues.

Even though DAB is much more sensitive chromogen compared to amino ethyl carbazol (AEC) because of its carcinogenic nature, some labs avoid using DAB. To resolve this problem, we have designed an AEC chromogen in liquid format. AEC is a chromogen of choice when performing immunoperoxidase staining. It has been well accepted amongst the pathologist because of its relatively less toxic nature compared to DAP. It produces a red colored end product at the positive sites which gives a good contrast with blue hematoxylin counter stain. Specimens stained in AEC cannot be dehydrated in ethanol and hence should be mounted in aqueous based mounting medium.
