Description:   Adrenergic receptors mediate the hormone and neurotransmitter functions of adrenaline and noradrenaline. Adrenoreceptors function in peripheral tissues and the central nervous system to regulate numerous vascular, metabolic, and neurological functions, and are the target for many therapeutically relevant drugs including those for the treatment of hypertension, asthma, benign prostatic hypertrophy, and depression. Adrenoreceptors are classified by three types (α1, α2, and β) and multiple subclasses based on pharmacological and molecular characteristics. The α1 family consists of three receptor subtypes (α1a, 1b, 1d) which primarily signal via Gq/11 alpha subunits to stimulate Phospholipase C and increase intracellular calcium. In contrast, the three α2 family receptor subtypes (α2a, 2b, 2c) signal via Gi/o alpha subunits to inhibit adenylyl cyclase activity, increase potassium transport, inhibit calcium conductance, and stimulate phospholipase A2. α1b adrenergic receptor mRNA expression in human tissues is highest in the spleen, kidney, and fetal brain. The α2a receptor is most highly expressed in human brain, spleen, and kidney, and is a species orthologue of the α2d receptor expressed similarly in rat, mouse, and cow.

Description:   The β-amyloid peptide (β A4), proteolytically released from the amyloid precursor protein (APP), provides the principal component of senile plaques in Alzheimer's disease. Cleavage of APP by α-secretase or alternatively by β-secretase leads to generation and extracellular release of soluble APP peptides, S-APP-α and S-APP-β, respectively, and the retention of corresponding membrane-anchored C-terminal fragments, C83 and C99. Subsequent processing of C83 by γ-secretase yields P3 peptides. This is the major secretory pathway and is nonamyloidogenic. Alternatively, presenilin/ nicastrin-mediated γ-secretase processing of C99 releases the amyloid β proteins, amyloid-β 40 (Aβ40) and amyloid-β 42 (Aβ42), major components of amyloid plaques, and the cytotoxic C-terminal fragments, γ-CTF(50), γ-CTF(57) and γ-CTF(59).

Description:   Dopamine is the predominant catecholamine neurotransmitter in the mammalian brain, where it regulates locomotor, emotional, memory, and dietary intake functions, as well as peripheral roles such as hormone secretion and renal function. Dopamine receptors (D1-D5) are members of the seven transmembrane domain containing G-protein coupled receptor (GPCR) family A, and are the target of several anti-psychosis and anti-Parkinsonian drugs. The D2, D3, and D4 dopamine receptors are encoded by homologous intron-containing genes, and signal via coupling of Gi/Go alpha G-protein subunits to inhibit adenylyl cyclase activity, as well as modulate cellular ion channel conductance of K+ and Ca2+. This is in contrast to D1 and D5 dopamine receptors, which signal via Gs alpha subunits to activate adenylyl cyclase activity. Coactivation of D1 and D2 receptors has been shown to activate a Gq mediated pathway, stimulating phospholipase C (PLC) and mobilizing intracellular calcium stores. D1 receptors were initially characterized in the striatum and retina and are expressed throughout the brain in the olfactory tubercle, islands of Calleja, entopeduncular nucleus, neocortex, hippocampus, and amygdala. Expression of the D2 receptor is found primarily in the brain (striatum, olfactory tubercle, and nucleus accumbens) and in the anterior and intermediate lobes of the pituitary.

Description:   The mTOR (mammalian target of rapamycin) pathway coordinates nutrient signals with growth factor dependent signaling. mTOR is organized in two independent protein complexes, mTORC1 and mTORC2. The mTORC1 complex containing mTOR, GβL/mLST8 and Raptor is sensitive to rapamycin, while the mTORC2 complex containing mTOR, GβL/mLST8, SIN1, and Rictor is rapamycin insensitive.
