Description:
CD24 is expressed in B cells until their terminal differentiation into plasma cells. It has been used widely as a marker to fractionate different B-lineage differentiation stages. CD24 has been found on a variety of other hematopoietic and neural cell types and is now classified as a costimulatory molecule. The CD24 molecule, also known as HSA, is composed mostly of carbohydrate with a protein core of only 31-35 amino acids in the human and 27 amino acids in the mouse. CD24 is now included in the large group of phosphatidylinositol-anchored membrane proteins. Studies with purified CD24 applied to latex beads have demonstrated a self-binding property suggesting that the molecule is capable of promoting homotypic aggregation, binding, and cell adhesion. This is supported by earlier observations of the blockade of B cell lymphoblast aggregation with an anti-CD24 monoclonal antibody. Recent studies of murine CD24 have assigned a role for CD24 as a ligand for P-selectin, a calcium-dependent lectin.

CD86, also known as B7-2, is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is expressed at high levels on resting peripheral monocytes and dendritic cells and at very low density on resting B and T lymphocytes. CD86 expression is rapidly upregulated by B-cell specific stimuli with peak expression at 18-42 hours after stimulation. CD86, along with CD80 (B7-1), is an important accessory molecule in T cell costimulation via its interaction with CD28 and CD152 (CTLA-4). Since CD86 has rapid kinetics of induction, it is believed to be the major CD28 ligand expressed early in the immune response. The monoclonal antibody GL1 blocks mixed lymphocyte reactions and stimulation of T cells by antigen-presenting cells.

CD44, also known as phagocytic glycoprotein-1 (Pgp-1), exists as a large number of different isoforms resulting from alternative RNA splicing. The major isoform expressed on lymphocytes, myeloid cells, and erythrocytes is a glycosylated type I transmembrane protein. Other variable isoforms containing glycosaminoglycans have molecular weights ranging from 110 to 250 kDa and are widely expressed on hematopoietic and non-hematopoietic cells. CD44 contributes to the adhesion of leukocytes to endothelial cells, stromal cells, and the extracellular matrix. It also plays a functional role in cell migration, lymphocyte homing, and adhesion during hematopoiesis and lymphocyte activation.

The fibrillins are extracellular matrix glycoproteins which occur as a major component of a subset of connective tissue microfibrils. These microfibrils have a beaded appearance and a cross-sectional diameter of 10-12 nm. In elastic tissues, these structures are thought to provide the scaffold onto which elastin is assembled to form elastic fibers, although their function in non-elastic tissues is unclear. There is an aberrant expression of fibrillin in Marfan syndrome. The epitope for the monoclonal antibody 11C1.3 has been found to be within exons 18-20 (a.a. 723-909).

CD11b, the integrin αM subunit, combines with CD18, the integrin β2 subunit, to form the integrin Mac-1, also known as complement receptor 3 (CR3), which mediates adhesion to C3bi and ICAM-1/CD54. Mac-1 is expressed at varying levels on granulocytes, macrophages, dendritic cells, NK cells, and B-1 cells in the peritoneal and pleural cavities. Mac-1 expression is rapidly up-regulated on neutrophils after activation and in the same time period that CD62L is shed from the cell surface. CD106, also known as VCAM-1, is an adhesion molecule and a major mediator of the inflammatory response. It is expressed on activated microvascular endothelial cells in response to signals arising from immune responses in infection, graft rejection, tumor recognition and killing. The complementary binding ligand for VCAM-1 is VLA-4/CD49d. In addition to VCAM-1, VLA-4 also recognizes the extracellular matrix molecule fibronectin. This pairing of VCAM-1 and VLA-4 is able to provide a second signal (e.g., non-antigen specific) for T cell stimulation, such as that seen in transplantation. The monoclonal antibody MK-2 has been used in transplant studies to suppress cardiac rejection and induce long-term cardiac graft survival. In addition to inflammatory responses, VCAM-1 has a significant role in hemopoiesis through its ability to retain lymphocyte and myeloid precursors on stromal cells in the marrow and lymphoid organs. CD106/VCAM-1 exists as an integral membrane protein. The M/K-2 monoclonal antibody immunoprecipitates a peptide that gives a single band on SDS-PAGE gels with an apparent Mr of ~100 kDa under reducing conditions and 92 kDa under non-reducing conditions.

CD31, also known as platelet endothelial cell adhesion molecule-1 (PECAM-1), is a type I integral membrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is constitutively expressed on the surface of endothelial cells and concentrated at the junction between them. It is also weakly expressed on many peripheral lymphoid cells and platelets. CD31 interacts homotypically in cell adhesion assays. The monoclonal antibody 390 inhibits the aggregation of L cells transfected with a variant form of CD31.
