The total protein is measured using the Biuret procedure, with bovine albumin as a standard. Antibody titer of the IgG fraction is standardized by equivalence-point precipitation. F(ab')2 fragments are tested for purity and specificity at a minimum of 20 mg/mL using immunoelectrophoresis. The product is goat F(ab')2; no trace of albumin, whole molecule IgG or Fc is detected.
The F(ab')2 fragment is prepared from the specific goat IgG fraction by limited digestion with pepsin, and purified by gel filtration or affinity chromatography. The resulting F(ab')2 fragment is dialyzed into 0.02M sodium phosphate, 0.14M sodium chloride, pH 7.3, 0.05% sodium azide, adjusted to standard titer, filtered through a 0.22 μm filter, vialed and lyophilized.
Goat F(AB')2 fragment to Human IgM is used as a precipitating antibody. It gives sharp precipitin lines in immunoelectrophoresis and immunodiffusion techniques. This product is suitable for conjugation with enzymes, radiolabels, fluorochromes or other markers, and for attachment to solid supports for use as an immunoadsorbant. This product is suitable for use as a primary reagent in enzyme immunoassays, fluorescein immunoassays and blot, cell or tissue immunostaining. F(ab')2 fragments are recommended for staining of cells or tissues which contain Fc receptors. (Note: Affinity purified antibodies are recommended to avoid non-specific binding from the inherent antibodies of host animals).

Erythrosin B Sodium Salt is used as a counter stain to hematoxylin, crystal violet and other nuclear stains, in both plant and animal tissue. Erythosin B dye solution in 0.9% saline is made for intravenous injection to induce occlusion.

1g dissolves in 0.9 mL water; 0.3 mL boiling water, 7.2 mL absolute alcohol, 4.2 mL methanol; insoluble in glycerol. The dissolution of sodium hydroxide in water is a highly exothermic reaction. Sodium hydroxide stock solutions should be stored in plastic containers. Glass containers should be avoided in the preparation and storage of sodium hydroxide solutions because it will etch the glass (typically anything over 2 N will etch glass). Keep solutions protected from air as much as possible since contact with air will form sodium carbonate.

DL-Dithiothreitol is also known as Clelands reagent; Protective agent for sulfhydryl groups (-SH). Quantitatively reduces disulfides (-S-S- to -SH). In this reaction the DTT is oxidized to the cyclic disulfide which ensures the reduction of other disulfides in solution. Disulfide reduction occurs quickly at pH 8. Dithiothreitol is useful for stabilizing sulfhydryl containing enzymes. Effective in sample buffers for reducing protein disulfide bonds prior to SDS-PAGE. DTT can also be used for reducing the disulfide bridge of the cross-linker N,N'-bis(acryloyl)cystamine to break apart the matrix of a polyacrylamide gel. DTT is less pungent and is less toxic than 2-mercaptoethanol. Useful for stabilizing sulfhydryl-containing enzymes.

β-Mercaptoethanol can act as an enzyme reactivator in systems necessitating reduction for activation, and has been commonly used to reduce disulfide bonds in order to separate protein subunits for use in electrophoresis. 2-Mercaptoethanol is used to reduce disulfide linkages in solubilizing proteins for gel electrophoresis (typically used in SDS-PAGE sample buffer at 5% concentration). Also reduces excess oxidative polymerization of catalysts. Cleaving intermolecular (between subunits) disulfide bonds allows the subunits of a protein to separate independently on SDS-PAGE. Cleaving intramolecular (within subunit) disulfide bonds allows the subunits to become completely denatured so that each peptide migrates according to its chain length with no influence due to secondary structure. In solution, 2-mercaptoethanol is readily oxidized in air to a disulfide, especially at alkaline pH. Because of this property, it is widely used to protect proteins, enzymes in particular, from becoming inactive. An excess of 2-mercaptoethanol (generally used at 0.01 M) will maintain the protein thiol groups in their reduced state.
