Bim belongs to the Bcl-2 protein family. Bcl-2 family members form hetero- or homodimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. This protein contains a Bcl-2 homology domain 3 (BH3). It has been shown to interact with other members of the Bcl-2 protein family (BCL2, BCL2L1/BCL-X(L) and MCL1) and to act as an apoptotic activator, inducing apoptosis and anoikis. Several variants have been identified. The isoforms vary in cytotoxicity with isoform BimS being the most potent and isoform BimEL being the least potent.

Interleukin-21 receptor (IL-21R) is a type I transmembrane glycoprotein within the class I cytokine receptor family, type 4 subfamily. Complex formation between IL-21R and the common gamma chain (gammac) is required for signaling. IL-21R is expressed mainly on B cells (highest on mature, activated, follicular and germinal center B cells), NK cells and activated T cells, but is also found on dendritic cells, alternatively activated macrophages, intestinal lamina propria fibroblasts and epithelial cells and keratinocytes. B cell IL-21R engagement induces Blimp1 (which mediates plasma cell differentiation) and is important for memory responses. IL-21R engagement on mouse NK cells enhances their cytotoxic activity and IFN-gamma production. IL-21R engagement on CD8+ T cells aids control of viral infection and tumor growth; IL-21R is also necessary for sufficient numbers of regulatory T cells to combat chronic inflammation. IL-21R expression is often upregulated in allergic skin inflammation, systemic lupus erythematosus and diffuse large B cell lymphoma (DLBCL).

The compound is a reversible inhibitor, and exhibits a linear mixed-type inhibition against ATP and peptide substrate. Alcian blue 8GX is primarily used for demonstrating acid mucopolysaccharides, which it does quite selectively. In recent years there has been some difficulty obtaining the dye. Alcian Blue 8GX is also suitable for detection of glycoproteins on nitrocellulose and in PAGE gels. There are other dyes similar to alcian blue, although not necessarily belonging to the same class of dyes. Alcian green has gained some popularity, and alcian yellow has been suggested, especially in a method for demonstrating Helicobacter. They are used for acid mucopolysaccharides in the same way as alcian blue. They have no particular advantage for demonstrating mucin. According to the Merck Index the groups marked X split off during staining.
