Description:
DnaJ-like proteins interact with HSP 70 molecular chaperones and function to facilitate protein folding and mitochondrial protein import. HSP 40-4, also known as HDJ2, is the human DnaJ homolog that functions as a co-chaperone with a cysteine-rich zinc finger domain. The cellular redox enzyme thioredoxin interacts with HSP 40-4, and oxidation and reduction reversibly regulate HSP 40-4 function in response to the changing redox states of the cell. The zinc finger domain of HSP 40-4 may act as a redox sensor of chaperone-mediated protein-folding machinery, since HSP 40-4 inactivation leads to the oxidation of cysteine thiols and a simultaneous release of coordinated zinc. Loss of the HSP 40-4 protein may be linked to severe defects in spermatogenesis that involve aberrant androgen signaling.

N-methyl-4-hydrazino-7-nitrobenzofurezan (MNBDH) is a new reagent for the determination of aldehydes and ketones. MNBDH reacts with carbonyl copmounds in acidic media to the corresponding MNBD-hydrazones. For example formaldehyde hydrazone formed is determined by high-performance liquid chromatography with UV/visible detection at 474 nm. MNBDH has been studied recently as a reagent with reduced interferences by ozone and nitrogen dioxide. It reacts with these oxidants to give only one product N-methyl-4-amino-7-nitrobenzofurezan (MNBDA).

ALKB protects DNA and RNA against damage from methylating compounds from the environment by directly reversing 1-methyladenine (1-meA) and 3-methylcytosine (3-meC) cytotoxic alkylation lesions in DNA and RNA. The enzymes act by oxidative demethylation, utilizing ferrous iron and alpha-ketoglutarate as cofactors, 2-oxoglutarate as a co-substrate, and molecular oxygen as the oxidizing agent. Deficiencies in DNA and RNA repair in mammals are associated with cancer, neurological disease and developmental defects. ALKB plays a role in resistance to anti-cancer drugs which attempt to damage tumor DNA. Escherichia coli ALKB protein belongs to the superfamily of 2-oxoglutarate- and iron(II)-dependent oxygenases.

Efficiently catalyzes both glutathionylation and deglutathionylation of mitochondrial complex I, which in turn regulates the superoxide production by the complex. Overexpression decreases the susceptibility to apoptosis and prevents loss of cardiolipin and cytochrome c release.

Ferritin heavy chains catalyze the first step in iron storage, the oxidation of Fe (II), whereas ferritin light chains promote the nucleation of ferrihydrite, enabling storage of Fe (III). Light chain ferritin is involved in cataracts by at least two mechanisms, hereditary hyperferritinemia cataract syndrome, in which light chain ferritin is overexpressed, and oxidative stress, an important factor in the development of ageing-related cataracts.
