Description:
SOD1 encoded by this gene binds copper and zinc ions and is one of two isozymes responsible for destroying free superoxide radicals in the body. The encoded isozyme is a soluble cytoplasmic protein, acting as a homodimer to convert naturally-occuring but harmful superoxide radicals to molecular oxygen and hydrogen peroxide. The other isozyme is a mitochondrial protein. Mutations in this gene have been implicated as causes of familial amyotrophic lateral sclerosis. Rare transcript variants have been reported for this gene. Mutations in the copper/zinc superoxide dismutase (SOD1) gene are associated with 15 to 20% of the familial forms of motoneuron disease. This 153 amino acid metalloenzyme is expressed in virtually all cells of all organisms above bacteria and is highly conserved across species, although some minor variations do occur. The incorporation of the mutated form of the SOD1 human gene into a transgenic mouse leads to the onset of the disease that closely resembles the human condition. These animals become weak at about 2 to 4 months of age and rapidly lose function, which results in death 4 to 6 weeks later.

Human galanin, a 30-amino acid non-amidated peptide, has originally been isolated from colon and pituitary. Its sequence differs from that of rat and porcine galanins by 4 and 6 amino acids in the C-terminal moiety, respectively. Synthetic human galanin is equipotent to the rat peptide in the isolated rat fundus muscle strip bioassay. Both peptides also fully activate the rat galanin receptor, indicating the importance of the conserved N-terminal portion of galanin for receptor interaction. Galanin, which is widely distributed in the CNS, and GAL receptors (GALR) are overexpressed in limbic brain regions associated with cognition in Alzheimer disease. As galanin inhibits acetylcholine release, galanin antagonists have gained interest in AD research.

CART (Cocaine- and Amphetamine-Regulated Transcript) is a recently discovered peptide, which was initially identified as mRNA produced primarily in the rat hypothalamus after administration of psychomotor stimulants. It is a satiety factor and closely associated with the actions of two important regulators of food intake, leptin and neuropeptide Y. When systemic leptin or receptors for leptin are inhibited, expression of CART mRNA is suppressed. CART (55-102) has been isolated from the hypothalamus of ovine suggesting in vivo processing at the Lys⁵³-Arg⁵⁴ region. When injected intracerebroventricularly into rats, recombinant CART peptides inhibited both normal and starvation-induced feeding, and completely blocked the feeding response induced by neuropeptide Y. Thus far, CART (55-102) appears to be the most potent fragment with a conserved secondary structure consisting of three disulfide bridges. Disruption of this secondary structure by reduction leads to a loss of the appetite induction.

Small synthetic molecules called growth-hormone secretagogues (GHSs) stimulate the release of growth hormone (GH) from the pituitary through a G protein-coupled receptor (GHS-R). Recently, ghrelin ("ghre" is the Proto-Indo-European root of the word "grow") has been identified as an endogenous ligand for GHS-R. These peptides consist of 28 amino acids with an octanoyl-residue at Ser³, which is essential for expressing activity. Ghrelin stimulates GH release from rat primary culture pituitary cells in a dose-dependent manner and induces an increase in intracellular Ca²⁺ in GHS-R-expressing cells with an EC₅₀ value of 2.5 nM. Human ghrelin is homologous to rat ghrelin apart from two amino acids. The occurrence of ghrelin in both rat and human indicates that GH release from the pituitary may be regulated not only by hypothalamic GHRH, but also by ghrelin. In future, ghrelin might be used in the treatment of aging patients as well as in patients with idiopathic GH deficiency, obesity, osteoporosis or cardiovascular diseases.
