Description:   Acts as part of the IKK complex in the conventional pathway of NF-kappa-B activation and phosphorylates inhibitors of NF-kappa-B thus leading to the dissociation of the inhibitor/NF-kappa-B complex and ultimately the degradation of the inhibitor. As part of the non-canonical pathway of NF-kappa-B activation, the MAP3K14-activated CHUK/IKKA homodimer phosphorylates NFKB2/p100 associated with RelB, inducing its proteolytic processing to NFKB2/p52 and the formation of NF-kappa-B RelB-p52 complexes. Also phosphorylates NCOA3.

Description:   TAK1 (or MAP3K7) was shown to participate in regulation of transcription by transforming growth factor beta (TGF beta). TAK1 is stimulated in response to TGF beta and bone morphogenetic protein. These results suggest that TAK1 functions as a mediator in the signaling pathway of TGF beta superfamily members. TAB1 and TAB2 are TAK1 binding proteins that may function as activators of the TAK1 (TGF b activated kinase 1) MAPKKK in TGF b signal transduction. TAB1 induced TAK1 activation promoted the dissociation of active forms of IKKa and IKK b from active TAK1, whereas the IKK mutants remained to interact with active TAK1. TNF a activated endogenous TAK1, and the kinase negative TAK1 acted as a dominant negative inhibitor against TNF a induced NFkB activation. TAK1 was suggested to act as a regulatory kinase of IKKs.

Description:   Serine kinase that plays an essential role in the NF-kappa-B signaling pathway which is activated by multiple stimuli such as inflammatory cytokines, bacterial or viral products, DNA damages or other cellular stresses. Acts as part of the canonical IKK complex in the conventional pathway of NF-kappa-B activation and phosphorylates inhibitors of NF-kappa-B on 2 critical serine residues. These modifications allow polyubiquitination of the inhibitors and subsequent degradation by the proteasome. In turn, free NF-kappa-B is translocated into the nucleus and activates the transcription of hundreds of genes involved in immune response, growth control, or protection against apoptosis. In addition to the NF-kappa-B inhibitors, phosphorylates several other components of the signaling pathway including NEMO/IKBKG, NF-kappa-B subunits RELA and NFKB1, as well as IKK-related kinases TBK1 and IKBKE. IKK-related kinase phosphorylations may prevent the overproduction of inflammatory mediators since they exert a negative regulation on canonical IKKs. Also phosphorylates other substrates including NCOA3, BCL10 and IRS1. Within the nucleus, acts as an adapter protein for NFKBIA degradation in UV-induced NF-kappa-B activation.

Description:   The CUE (coupling of ubiquitin conjugation to endoplasmic reticulum degradation) domain is an evolutionarily conserved, ~40 amino acid monoubiquitin-binding domain that mediates intramolecular monoubiquitylation. CUE domains are present in eukaryotic proteins that are involved in ubiquitination and protein trafficking pathways and may be required for ubiquitination of the proteins in which they are found. CUEDC2 (CUE domain-containing protein 2) was found through a yeast two-hybrid screening as a protein that interacts with the progesterone receptor (PR) and promotes progesterone-induced PR degradation by the ubiquitin-proteasome pathway. CUEDC2 also decreases the sumoylation of PR. CUEDC2 has also been found to interact with IKK-alpha and IKK-beta and decrease the activation of NF-kB by decreasing the activation of IKK.

Description:   Anti-NEMO antibody was designed, produced, and validated as part of the Joy Cappel Young Investigator Award (JCYIA). Anti-NEMO antibody detects recombinant and endogenous NEMO. NEMO, the regulatory subunit of the IKK core complex, phosphorylates inhibitors of NF-kappa-B thus leading to the dissociation of the inhibitor/NF-kappa-B complex and ultimately the degradation of the inhibitor. Its binding to scaffolding polyubiquitin seems to play a role in IKK activation by multiple signaling receptor pathways. Nemo is also considered to be a mediator for TAX activation of NF-kappa-B and may be implicated in NF-kappa-B-mediated protection from cytokine toxicity. NEMO is essential for viral activation of IRF3 and involved in TLR3- and IFIH1-mediated antiviral innate response. The innate antivaral response from NEMO requires 'Lys-27'-linked polyubiquitination. Anti-NEMO is ideal for researchers interested in Immunology and Cancer research.
