Description: This antibody recognizes a protein of 25-26kDa, identified as bcl-2 alpha oncoprotein. It shows no cross-reaction with BCL-X or BAX protein. Expression of BCL-2 alpha oncoprotein inhibits the programmed cell death (apoptosis). In most follicular lymphomas, neoplastic germinal centers express high levels of BCL-2 alpha protein, whereas the normal or hyperplastic germinal centers are negative. This antibody is valuable when distinguishing between reactive and neoplastic follicular proliferation in lymph node biopsies. It may also be used in distinguishing between those follicular lymphomas that express BCL-2 protein and small number in which neoplastic cells are BCL-2 negative.

Description: The HMB45 clone reacts with a neuraminidase-sensitive oligosaccharide side chain of a glycoconjugate present in immature melanosomes. The HMB45-reactive antigen is present in cutaneous melanocytes, prenatal and infantile retinal pigment epithelium and melanoma cells and is thought to be oncofetal in nature. This antibody has been shown to label the majority of melanomas. Clone A103 recognizes a protein of 20 kDa, identified as MART-1 (melanoma antigen recognized by T cells-1) or Melan-A. Melan-A is a useful addition to melanoma panels as it is apparently specific for melanocytic lesions. Studies have also shown that MART-1 is more sensitive than HMB45 when labeling metastatic melanomas. Tyrosinase is a key enzyme involved in the initial stages of melanin biosynthesis. Studies have shown tyrosinase to be a more sensitive marker when compared to HMB45 and MART-1. It has been shown to label a higher percentage of desmoplastic melanomas than HMB45. The combination of HMB45, MART-1 and tyrosinase make this triple antibody cocktail a first-order pan melanoma screener.

Description: SMADs are members of the MAD-related family of molecules. MAD-related proteins are a family of intracellular proteins that are essential components in the signaling pathways of the serine/threonine kinase receptors of the transforming growth factor beta superfamily. Alterations in the SMAD4 gene was primarily discovered in pancreatic cancer (duct adenocarcinoma) but occur in a variety of cancers such as colorectal cancer, gastric cancer, prostate cancer, melanomas, head and neck cancers and many others, though with higher frequencies in gastrointestinal tract cancers. Loss of SMAD4 expression in tumors has also been shown to affect cancer progression and therapy, such as reduced response to adjuvant chemotherapy.

Description: Anti-Cytokeratin (CAM 5.2) reagent has a primary reactivity with human keratin proteins that correspond to Moll’s peptides #7 and #8, Mr 48 and 52 kilodaltons (kd), respectively. Cytokeratin 7 and 8 are present on secretory epithelia of normal human tissue but not onstratified squamous epithelium. Anti-Cytokeratin (CAM 5.2) stains most epithelial-derived tissue, including liver, renal tubular epithelium, and hepatocellular and renal cell carcinomas.
