Description:   Human CD166 is a type 1 glycoprotein expressed on activated T cells, B cells and monocytes and appears to be the ligand for CD6. Human CD166 may be important for activation of T cells.

Description:   Human CD152 (CTLA-4) is a cell surface glycoprotein expressed at low levels on activated T cells. CD152 is a high affinity receptor for the costimulatory molecules CD80 (B7-1) and CD86 (B7-2) and appears to function as a negative regulator of T cell activation.

Description:   Interleukin-21 (IL-21) is a key factor in the transition between innate and adaptive immune responses secreted by activated T cells. The IL-21 receptor (IL-21R) is expressed in lymphoid tissue, in particular by NK, B, T and dendritic cells, macrophages and endothelial cells. Recent evidence suggests that IL-21 plays a supportive role in the proliferation of T and B cells and influences the cytolytic activity of natural killer cells. IL-21 has been shown to up-regulate genes associated with innate immunity and to inhibit the differentiation of naïve T helper cells. IL-21 specifically inhibits IFN-gamma production from developing TH1 cells and is preferentially expressed by TH2 cells. Furthermore IL-21 has been identified as a growth and survival factor for human myeloma cells. IL-21/IL-21R interactions have a unique role in sequentially activating both innate and adaptive immune responses against poorly immunogenic tumors, leading to tumor rejection that is perforin dependent but IFN-gamma independent.

Description:   Human CD45 is expressed on all cells of hematopoietic origin, except erythrocytes. CD45 is a transmembrane tyrosine phosphatase which can exist in at least nine different isoforms resulting from tissue-specific alternative RNA splicing of exons 4-7 of a single gene coding for the various N-terminal peptide segments. The CD45RO isoform contains none of the alternatively spliced exons and is present on most thymocytes, a subpopulation of resting (memory) T cells within both CD4 and CD8 subsets as well as on monocytes and granulocytes. CD45RO plays an essential role in lymphocyte activation.

Description:   Human CD272 (BTLA; B and T Lymphocyte Attenuator) is a member of the immunoglobulin superfamily and has sequence homology to PD-1 and CTLA-4. It is expressed on T and B lymphocytes and other hemopoetic lineages. Engagement of this molecule by its ligand CD270 (HVEM) can downregulate activated T and B cell responses. CD272 levels on antigen specific CD8+ T cells have been reported to decrease in viral specific, but not melanoma specific activated lines. Polymorphism of the CD272 molecule has been linked to rheumatoid arthritis.

Description:   The transmembrane and secreted protein delta-like 1 homolog (DLK1; Protein delta homolog 1; Preadipocyte factor 1; Pref-1) belongs to the EGF-like family. It can be cleaved by a tumor necrosis factor-alpha converting enzyme (TACE) to release a soluble form. DLK1 plays important roles in regulating cell differentiation such as adipogenesis and osteogenesis. Membrane-bound DLK1 promotes hypertrophic myotube formation, whereas soluble DLK1 inhibits myotube formation. The soluble form of DLK1 inhibits preadipocyte differentiation, while membrane DLK1 represses preadipocyte proliferation. DLK1 is also associated with tumor invasion.

Description:   The p75 low affinity human Nerve Growth Factor receptor CD271(NGRF, TNFRSF16) is expressed on a variety of tissue types including a subset of splenic and nodal lymphocytes. Presence of this receptor supports uptake of intracellular calcium, but not mobilization. CD271 is a stem cell marker for hematopoietic, stromal as well as cancer cells. CD271 has been reported to inhibit differentiation of mesenchymal cells into various lineages.

Description:   The TNF family ligand ectodysplasin A (EDA) and its receptor EDAR are required for proper development of skin appendages such as hair, teeth and eccrine sweat glands. Loss of function mutations in the Eda gene cause X-linked hypohidrotic ectodermal dysplasia (XLHED), a condition that can be ameliorated in mice and dogs by timely administration of recombinant EDA. The Eda gene on the X chromosome is transcribed as multiple splice variants, only two of which code for the receptor-binding C-terminal TNF homology domain. These two variants code for 391- and 389-amino acid-long proteins called EDA1 and EDA2. EDA1 binds EDAR, whereas EDA2 binds to another receptor, XEDAR. The biology of EDA2 and XEDAR is distinct from that of EDA1. Indeed, XEDAR-deficient mice have no obvious ectodermal dysplasia phenotype, whereas mice deficient in EDA, EDAR, or the signaling adaptor protein EDARADD all display virtually indistinguishable ectodermal dysplasia phenotypes, indicating the predominance of the EDA1-EDAR axis in the development of skin-derived appendages.
