Description:
Nuclear factor kappa B (NF-kB) is a ubiquitous transcription factor and an essential mediator of gene expression during activation of immune and inflammatory responses. NF-kB mediates the expression of a great variety of genes in response to extracellular stimuli including IL-1, TNFa and bacteria product LPS. NF-kB is associated with IkB proteins in the cell cytoplasm, which inhibit NF-kB activity. The long-sought IkB kinase (IKK), which phosphorylates IkB, and mediates IkB degradation and NF-kB activation, was recently identified by several laboratories. IKK is a serine protein kinase, and the IKK complex contains alpha and beta subunits (IKKa and IKKb). IKKa and IKKb interact with each other and both are essential for the NF-kB activation. IKKa specifically phosphorylates IkB-alpha. IKKa is expressed in a variety of human tissues.

Phosphorylation of I-kB by IkB kinase (IKK) complex leads to degradation of I-kB and activation of NF-kB. The IKK complex contains IKKa,IKKb, and IKKg. A novel IKK related kinase was recently identified and designated TBK1 (TANK-binding kinase 1), NAK (NF-kB-activating kinase), and T2K. NAK/TBK1 activates IKKb through direct phosphorylation. NAK/TBK1is activated by growth factors and PMA and mediates IKK and NF-kB activation in response to growth factors. NAK/TBK1 functions upstream of NIK and the IKK complex. NAK/TBK1 is also critical in protecting embryonic liver from apoptosis.

Inhibitor of kappa light polypeptide gene enhancer in B-cells, kinase epsilon.The transcription factor NFκB is retained in the cytoplasm in an inactive form by the inhibitory protein IκB. Activation of NFκB requires that IκB be phosphorylated on specific serine residues, which results in targeted degra-dation of IκB. IκB kinase beta(IKKbeta), previously designated CHUK, interacts with IκB-betaand specifically phosphorylates IκB-betaon the sites that trigger its degradation, serines 32 and 36. The functional IKK complex contains three subunits, IKKbeta, IKKbetaand IKKgamma(also designated NEMO), and each appear to make essential contributions to IκB phosphorylation. IKK-i is a serine/threonine kinase that shares homology with IKKbetaand IKKbeta. IKK-i is pri-marily expressed in immune cells and is induced by lipopolysaccharide and by proinflammatory cytokines including TNFbeta, IL-1 and IL-6. Overexpression of IKK-i was shown to result in phosphorylation of IκBbetaon Ser32 and Ser36, and in NFκB activation, suggesting that IKK-i may act as an IκB kinase in the immune system.
