Microtubules are key elements of the eukaryotic cytoskeleton that dynamically assemble from heterodimers of alpha- and beta-tubulin. Two different mechanisms can generate microtubule diversity: the expression of different alpha- and beta-tubulin genes, referred to as tubulin isotypes, and the generation of posttranslational modifications (PTMs) on alpha- and beta-tubulin. Tubulin PTMs include the well-known acetylation or phosphorylation, and others that have so far mostly been found on tubulin, detyrosination/tyrosination, polyglutamylation and polyglycylation. These PTMs might have evolved to specifically regulate tubulin and microtubule functions. Polyglutamylation is a PTM that occurs when secondary glutamate side chains are formed on gamma-carboxyl groups of glutamate residues in a protein. Enzymes catalyzing polyglutamylation belong to the TTL-like (TTLL; Tubulin tyrosine ligase-like) family of glutamylases. Deglutamylases, the enzymes that reverse polyglutamylation, were identified within a novel family of CCPs (cytosolic carboxypeptidase). Subtle differences in polyglutamylation can be seen on diverse microtubules in different cell types. The functions of these modifications remain to be studied. However, its wide distribution strengthens the idea that it could be involved in fine-tuning a range of microtubule functions.

Clusterin shares homology with the small heat shock protein family of molecular chaperones. The mature secreted form of the protein is a glycosylated, 80-kDa disulfide-linked heterodimer of alpha and beta subunits (produced by internal cleavage). Clusterin is expressed in virtually all tissues and found in all human fluids. It is involved in numerous physiological processes important for carcinogenesis and tumor growth, including apoptotic cell death, cell cycle regulation, DNA repair, cell adhesion, tissue remodeling, lipid transportation, membrane recycling, and immune system regulation. Clusterin also exists as a nuclear protein. The secreted form of Clusterin has extracellular chaperone and anti-apoptotic activities while the nuclear form acts as a proapoptotic factor.

Betatrophin (RIFL; Lipasin; Angiopoietin-like protein 8 (ANGPTL8)) is a newly discovered secreted protein of 198 aa that was proposed to promote beta cell proliferation and improve glucose tolerance in mice. Betatrophin may also function in inhibition of lipase activity and on serum triglyceride regulation. Betatrophin is expressed in the liver and in white and brown adipose tissue of mice. In humans, betatrophin is found to be predominantly expressed in the liver. Betatrophin levels are reduced by fasting and are elevated upon insulin resistance and during pregnancy. Betatrophin, according to preliminary data could bind to the macrophage receptor Marco and also to RTN4R, a neuronal receptor. Recently, a study using ANGPTL8 KO mice showed that ANGPTL8/Betatrophin does not play a role in beta cell proliferation nor in glycemic control as previously thought, but regulates plasma triglyceride levels.
