Human CD83 is a 40-50 kDa member of the Siglec (or sialic-acid-binding immunoglobulin-like lectin) family of transmembrane proteins. CD83 is a primary marker for dendritic cells. It is also found on B cells, neutrophils, monocytes and macrophages. Except for dendritic cells, CD83 expression is often transient. CD83 binds to sialic acids on target cells. Membrane CD83 promotes T cell proliferation, particularly of CD8+ cytotoxic T cells. Soluble CD83 is immunosuppressive and blocks T cell activation. On monocytes, CD83 is suggested to drive monocytes into a fibrocyte phenotype. A lack of membrane-expressed CD83 leads to an unusual IL-4/ IL-10 producing CD4+ T cell phenotype.

This common fluorescent reagent often used in carbohydrate analysis, is also useful in fluorescence visualization of lysosomes in live cell lines in culture. This fluorophore that has the unusual environmentally sensitive property of increasing fluorescence in low pH environments. Comparison of staining patterns with the common LysoTracker™ or LysoSensor™ dyes indicated comparable staining, albeit using a higher concentration of NBD-piperazine (1μM versus 100nM for the Lyso-Dyes) but at a lower cost per assay. Also used for the determination of mono- and diisocyantes in air samples, and determination of lactate, hydroxybutyrate and other intermediates in body fluids. The piperazino moiety reacts readily with isocyantes to form the corresponding urea-derivatives. The derivatives can be detected by LC with UV or fluorescent-detection. Activation of cells by LPS is mediated by the Toll-like receptor 4 (TLR4). For optimal interaction with LPS, TLR4 requires association with myeloid differentiation protein 2 (MD-2). According to current consensus activation of TLR4 is preceded by the transfer of LPS to membrane-bound (m) or soluble (s) CD14 by LPS-binding protein (LBP). Re-form LPS and lipid A, but not S-form LPS, are capable of inducing TNF-alpha responses also in the absence of CD14. LPS, synthesized by most wild-type (WT) Gram-negative bacteria (S-form LPS), consists of three regions, the O-polysaccharide chain, which is made up of repeating oligosaccharide units, the core oligosaccharide and the lipid A, which harbors the endotoxic activity of the entire molecule. R-form LPS synthesized by the so-called rough (R) mutants of Gram-negative bacteria lacks the O-specific chain. Furthermore, the core-oligosaccharide may be present in different degrees of completion, depending on the class (Ra to Re) to which the mutant belongs. LPS are amphipathic molecules whose hydrophobicity decreases with increasing length of the sugar part. Based upon these differences, S- and R-form LPS show marked differences in the kinetics of their blood clearance and cellular uptake as well as in the ability to induce oxidative burst in human granulocytes and to activate the host complement system.

Human CD28 is an important costimulatory molecule found on all CD4+ T cells and on about half of the CD8+ T cells. T cell activities attributed to CD28 include prevention of anergy, induction of cytokine gene transcription, stabilization of cytokine mRNA and activation of CD8+ cytotoxic T lymphocytes. The ligands for CD28 identified as CD80 (B7-1) and CD86 (B7-2) are immunglobulin superfamily monomeric transmembrane glycoproteins of 60kDa and 80kDa respectively. Anti-CD28 antibodies have been shown to have a strong adjuvant effect when conjugated to an immunogen. Monovalency of the CD28 receptor for its natural ligands is essential to provide costimulation without inducing responses in the absense of TCR engagement.
