Murine CD79 is a disulfide-linked heterodimer composed of an α chain (CD79a) and a β chain (CD79b) that associates non-covalently with membrane immunoglobulin (Ig) to form the B cell receptor (BCR) complex. Its expression is restricted to B lymphocytes first appearing on the surface at the pro-B cell stage prior to productive Ig gene rearrangements and remaining through all stages of B-cell differentiation prior to plasma cells. It has been proposed that the CD79 complex on pro-B cell surfaces may function to induce early B-cell differentiation.

In the mouse, CD8 exists in two forms – (i) a CD8 heterodimer composed of an α chain (CD8α) and a β chain (CD8β); and (ii) a homodimer of two α chains. The heterodimer is found on the surface of essentially all thymocytes and the “suppressor/cytotoxic” subpopulation of mature T lymphocytes. Subsets of intestinal intraepithelial lymphocytes express CD8α without CD8β. It has been suggested that CD8+β- T cells mature extrathymically, while development of the CD8α+β+ population of T cells is thymus-dependent. CD8 acts as a coreceptor with MHC Class I-restricted T cell receptors in antigen recognition and positive selection of MHC class I-restricted CD8+ T cells. In vivo and in vitro treatment with the 53-6.7 monoclonal antibody effectively depletes CD8α+ cells. The 53-6.7 monoclonal antibody also blocks allogeneic help specific for class I MHC antigens and T cell responses to IL-2.

CD49d, a member of the integrin family of extracellular matrix and cell adhesion molecules, is the integrin α4 chain. CD49d associates noncovalently with CD29, the integrin β1 subunit, to form the α4β1 very late antigen-4 (VLA-4) heterodimer. Integrin α4 can also associate with integrin β7 to form the α4β7 heterodimer. It is expressed on most leukocytes with the possible exception of neutrophils. VLA-4 mediates leukocyte binding to CD106 (VCAM-1) and is involved in the migration of leukocytes from blood to tissue at sites of inflammation. VLA-4 can also provide a costimulatory signal to T cells for activation and proliferation. The α4β7 integrin heterodimer is expressed on a small subpopulation of thymocytes, most lymph node T and B lymphocytes, NK cells, and eosinophils. Integrin α4β7 also binds to fibronectin and VCAM-1.

CD45R, also known as B220, a member of the protein tyrosine phosphate family and a major cell surface glycoprotein, represents a restricted form of the CD45 family which primarily recognizes only cells of B-lineage from pro-B cell through mature B lymphocytes and prior to the availability of anti-CD19 monoclonal antibodies was commonly used as a pan B-cell marker. It also reacts with certain activated T cells as well as non-MHC-restricted lytically active lymphokine-activated killer (LAK) cells. In vivo administration of RA3-6B2 has been shown to affect differentiation of both T and B cells in normal mice and reduce the level of anti-DNA antibodies and lymphadenopathies in MRL/lpr mice.
