CD62L, also known as L-selectin, is a type I transmembrane glycoprotein with lectin-like and epidermal growth factor-like domains. It binds to sialylated oligosaccharide determinants on high endothelial venules (HEV) in peripheral lymph nodes. In the mouse, CD62L is expressed on most thymocytes and on peripheral leukocytes, including B and T lymphocytes, neutrophils, monocytes, and eosinophils. This member of the selectin family of cell adhesion molecules appears to be required for lymphocyte homing to peripheral lymph nodes and for leukocyte extravasation at sites of inflammation. Expression of CD62L on lymphocytes and neutrophils is rapidly down-regulated upon cell activation and the level of CD62L expression, along with other markers, distinguishes naive T cells from effector/memory T cells. The monoclonal antibody MEL-14 blocks in vitro binding of lymphocytes to peripheral lymph node HEV and inhibits in vivo lymphocyte extravasation into peripheral lymph nodes and the late stages of leukocyte rolling.

Interleukin-21 (IL-21) is a four helical bundle type I cytokine. The mature form of human IL-21 is a 131 amino acid protein. IL-21 is expressed primarily by NKT and CD4+ T cells, particularly the inflammatory Th17 subset, and has pleiotropic effects on both innate and adaptive immune responses. The effects of IL-21 include promotion of antibody production, NK cell differentiation, proliferation and differentiation of cells of monocyte and granulocyte lineages, costimulation of T cells, and augmentation of antitumor activity of CD8+ T cells. Studies have shown that IL-21 is also an autocrine cytokine that potently induces Th17 differentiation and suppresses Foxp3 expression, and serves as a target for treating inflammatory diseases.

The BC96 monoclonal antibody reacts with human CD25, the 55 kDa interleukin-2 receptor α chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. By itself, CD25 binds IL-2 only with low affinity. However, CD25 associates with CD122 (IL-2 receptor β chain) and CD132 (common γ chain) to form the high affinity IL-2 receptor. CD25 plays a role in lymphocyte differentiating and activation/proliferation. CD25 is expressed on activated B cells, monocytes/macrophages, and T cells.
