Description:
CD3ε, a member of the immunoglobulin superfamily of cell surface receptors, is comprised of five invariable chains ranging in size from 16-28 kDa and is closely associated with the T cell antigen receptor (TCR). It is expressed on all T cells of all mouse strains. CD3 plays a major role in signaling during antigen recognition, leading to T-cell activation. The 145-2C11 monoclonal antibody recognizes an epitope on the 25 kDa ε chain of the CD3/TCR complex. In the presence of Fc receptor-bearing accessory cells, soluble 145-2C11 can activate primed and naïve T cell in vitro. 145-2C11 can also induce redirected lysis of specific target cells by CTL clones and it can block lysis of specific target cells by antigen-specific CTL’s. Immobilized 145-2C11 can activate both normal T lymphocytes and cloned T cell lines. Under certain conditions, T cell activation by 145-2C11 may result in apoptotic cell death.

The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. Expression of CD25 on activated T lymphocytes is transitory and endogenously regulated. CD25 is expressed on precursor B cells in bone marrow. Its expression is initiated by functional rearrangement and expression of IgM heavy chain genes and is down-regulated when immature B cells mature and express IgD. It is expressed at a higher level on CD4+CD8+ thymocytes. It is also expressed on cultured epidermal Langerhans cells. The biochemical features of murine CD25 have been characterized in detail. The 3C7 monoclonal antibody reacts with an epitope of CD25 which is distinct from that recognized by clone 7D4. 3C7, used in combination with 7D4 in culture, results in higher levels of inhibition of proliferation driven by IL-2 and generation of alloreactive CTL than either of these monoclonal antibodies alone.

CD44 represents a family of type I transmembrane glycoproteins expressed on many cell types such as B cells, monocytes, macrophages, and variable subsets of thymocytes and peripheral T cells. CD44 primarily functions as an adhesion molecule. It has been implicated in various processes, such as hematopoiesis, lymphocyte homing, leukocyte activation, tumor metastasis and development. The KM201 monoclonal antibody reacts with an epitope very close to the hyaluronate binding domain on the CD44 molecule. KM201 can inhibit hyaluronate-dependent cell aggregation, prevent lympho-hemopoiesis in both Dexter and Whitlock-Witte cultures, prevent the earliest intrathymic precursors from homing to the thymus, and costimulate the activation of freshly purified splenic CD4+ T cells.

CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

CD11b, the integrin αM subunit, combines with CD18, the integrin β2 subunit, to form the integrin Mac-1, also known as complement receptor 3 (CR3), which mediates adhesion to C3bi and ICAM-1/CD54. Mac-1 is expressed at varying levels on granulocytes, macrophages, dendritic cells, NK cells, and B-1 cells in the peritoneal and pleural cavities. Mac-1 expression is rapidly up-regulated on neutrophils after activation and in the same time period that CD62L is shed from the cell surface. In addition to its role in binding C3bi on opsonized targets and mediation of the subsequent ingestion process, Mac-1 is important as an adhesion molecule in the transendothelial migration of monocytes and neutrophils. Cross-reaction of M1/70 to human CD11b has been observed on monocytes, polymorphonuclear leukocytes, and NK cells.

The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosporylate the Src family of protein kinases. This activity is known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685 and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody 104 recognizes CD45 (leukocyte common antigen) on all leukocytes of mouse strains expressing the CD45.2 allotype (e.g., A, AKR, BALB/c, CBA/Ca, CBA/J, C3H/He, C57BL, C57BR, C57L, C58, DBA/1, DBA/2, NZB, SWR, 129).

Description:
The CD23 antigen is the low affinity IgE Fc receptor, which is a 45-49 kDa protein with 38 and 28 kDa fragments. It is expressed on most mature, conventional B cells (but not on peritoneal CD5+ B cells) and can also be found on the surface of T cells, macrophages and platelets. It is distinct from the high affinity IgE receptors found on basophils and mast cells which mediate allergic reactions. The low affinity receptors are thought to play a role in isotype specific immunoregulation. The regulation of CD23 surface expression appears to be integral with the complex IgE system which involves interactions of cells, cytokines, antibodies and regulatory factors. CD23 has been described as a “membrane bound cytokine," in that the soluble cleavage products of CD23 are themselves able to act as cytokines in vitro.
