The monoclonal antibody I21/7 reacts with the α subunit of mouse and human CD11a, also known as LFA-1, and a member of the integrin family of cell adhesion molecules. The antigen is expressed on thymocytes, T and B lymphocytes, and on non-lymphoid cells in the bone marrow. CD11a appears to be an essential component for T-cell activation by antigen-presenting cells and is important for cell-mediated killing. Costimulatory signaling by LFA-1 is achieved by pairing with its ligand, CD54/ICAM-1.

Description:
CD24, also known as heat stable antigen, is a phosphatidylinositol-linked cell surface glycoprotein that is expressed at all stages of B cell development and on most thymocytes. The absence of expression from mature T cells is closely associated with their maturation from CD4+CD8+CD24+ thymocytes to either CD4+CD8-CD24- or CD4-CD8+CD24- T cells. CD24 is also present on mouse granulocytes, monocytes, Langerhans cells and erythrocytes. Its ligand is P-selectin (CD62P). CD24 may play a role in regulation of B cell proliferation and differentiation, and it has been proposed that heat stable antigen promotes homotypic adhesion between B lymphocytes.

Description:
The CD23 antigen is the low affinity IgE Fc receptor, which is a 45-49 kDa protein with 38 and 28 kDa fragments. It is expressed on most mature, conventional B cells (but not on peritoneal CD5+ B cells) and can also be found on the surface of T cells, macrophages and platelets. It is distinct from the high affinity IgE receptors found on basophils and mast cells which mediate allergic reactions. The low affinity receptors are thought to play a role in isotype specific immunoregulation. The regulation of CD23 surface expression appears to be integral with the complex IgE system which involves interactions of cells, cytokines, antibodies and regulatory factors. CD23 has been described as a “membrane bound cytokine," in that the soluble cleavage products of CD23 are themselves able to act as cytokines in vitro.

Description:
CD62L, also known as L-selectin, is a type I transmembrane glycoprotein with lectin-like and epidermal growth factor-like domains. It binds to sialylated oligosaccharide determinants on high endothelial venules (HEV) in peripheral lymph nodes. In the mouse, CD62L is expressed on most thymocytes and on peripheral leukocytes, including B and T lymphocytes, neutrophils, monocytes, and eosinophils. This member of the selectin family of cell adhesion molecules appears to be required for lymphocyte homing to peripheral lymph nodes and for leukocyte extravasation at sites of inflammation. Expression of CD62L on lymphocytes and neutrophils is rapidly down-regulated upon cell activation and the level of CD62L expression, along with other markers, distinguishes naive T cells from effector/memory T cells. The monoclonal antibody MEL-14 blocks in vitro binding of lymphocytes to peripheral lymph node HEV and inhibits in vivo lymphocyte extravasation into peripheral lymph nodes and the late stages of leukocyte rolling.

The monoclonal antibody 18d3 reacts with the mouse CD94 receptor subunit, a type II transmembrane glycoprotein. CD94 is expressed on the surface of NK cells, NK T cells, and some memory phenotype CD8+ cells. When CD94 is paired with NKG2A, C, or E subunit, it binds the nonclassical MHC class Ib molecule Qa-1b and results in a CD94high population by flow cytometry. Approximately half of NK cells do not express NKG2A, C or E; these cells express CD94 on the surface in a form that does not bind Qa-1b and is believed to be nonfunctional, producing a CD94low population by flow cytometry. CD94 is thought to play a role in the adhesion and activation of the NK cell lineage.

CD22, also known as Lyb-8, is a heterodimeric transmembrane glycoprotein and a member of a structurally related group of immunoglobulin (Ig) superfamily domain-containing proteins called the sialoadhesin family. It is detected in the cytoplasm early in B cell development (late pro-B cell stage), appears on the cell surface concomitant with IgD, and is found on most IgM+IgD+ mature B lymphocytes. Expression is lost with terminal differentiation of B cells and is absent on plasma cells. Activation of B cells via cross-linking of surface Ig increases CD22 expression. CD22 associates with the B cell receptor (BCR) complex and mediates intercellular adhesion. Its intracellular domain is phosphorylated after antigen receptor cross-linking and is involved in negative regulation of B cell activation.

The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosphorylate the Src family of protein kinases. This activity is now known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685, and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The former arises as a result of alternative splicing of different exons. The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody A20 recognizes all mouse leukocytes expressing the CD45.1 allotype.
