ApoScreen® Annexin V (SB Cat. No. 10040) is a recombinant protein that is expressed in E. coli. A number of changes in cell structure occur during apoptosis. One major change is the loss of membrane phospholipid asymmetry, with translocation of phosphatidylserine (PS) from the inner leaflet of the phospholipid bilayer to the external surface. Annexin V, a member of the annexin family of calcium-dependent phospholipid binding proteins, has a high affinity for phosphatidylserine (PS)-containing phospholipid bilayers. Conjugated Annexin V can be used to monitor changes in cell membrane phospholipid asymmetry, thereby providing a convenient tool for detection of apoptotic cells.

Description:
In the mouse, CD8 exists in two forms – (i) a CD8 heterodimer composed of an α chain (CD8α) and a β chain (CD8β); and (ii) a homodimer of two α chains. The heterodimer is found on the surface of essentially all thymocytes and the “suppressor/cytotoxic” subpopulation of mature T lymphocytes. Subsets of intestinal intraepithelial lymphocytes express CD8α without CD8β. It has been suggested that CD8+β- T cells mature extrathymically, while development of the CD8α+β+ population of T cells is thymus-dependent. CD8 acts as a coreceptor with MHC Class I-restricted T cell receptors in antigen recognition and positive selection of MHC class I-restricted CD8+ T cells. In vivo and in vitro treatment with the 53-6.7 monoclonal antibody effectively depletes CD8α+ cells. The 53-6.7 monoclonal antibody also blocks allogeneic help specific for class I MHC antigens and T cell responses to IL-2.

Interleukin-21 (IL-21) is a four helical bundle type I cytokine. The mature form of human IL-21 is a 131 amino acid protein. IL-21 is expressed primarily by NKT and CD4+ T cells, particularly the inflammatory Th17 subset, and has pleiotropic effects on both innate and adaptive immune responses. The effects of IL-21 include promotion of antibody production, NK cell differentiation, proliferation and differentiation of cells of monocyte and granulocyte lineages, costimulation of T cells, and augmentation of antitumor activity of CD8+ T cells. Studies have shown that IL-21 is also an autocrine cytokine that potently induces Th17 differentiation and suppresses Foxp3 expression, and serves as a target for treating inflammatory diseases.

CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

CD4 is a 55 kDa type I transmembrane glycoprotein and a member of the Ig-SF of cell surface receptors. It is expressed predominantly on the “helper/inducer” subpopulation of mature T lymphocytes and on monocytes and macrophages. Domains 1 and 2 bind to MHC Class II antigens, while domains 3 and 4 may be involved in cis interactions with the CD3/TCR complex. CD4 functions as an accessory molecule in the recognition of foreign antigens in association with MHC class II antigens by T cells.
