Description:   The monoclonal antibody I21/7 reacts with the α subunit of mouse and human CD11a, also known as LFA-1, and a member of the integrin family of cell adhesion molecules. The antigen is expressed on thymocytes, T and B lymphocytes, and on non-lymphoid cells in the bone marrow. CD11a appears to be an essential component for T-cell activation by antigen-presenting cells and is important for cell-mediated killing. Costimulatory signaling by LFA-1 is achieved by pairing with its ligand, CD54/ICAM-1.

Description:   CD106, also known as VCAM-1, is an adhesion molecule and a major mediator of the inflammatory response. It is expressed on activated microvascular endothelial cells in response to signals arising from immune responses in infection, graft rejection, tumor recognition and killing. The complementary binding ligand for VCAM-1 is VLA-4/CD49d. In addition to VCAM-1, VLA-4 also recognizes the extracellular matrix molecule fibronectin. This pairing of VCAM-1 and VLA-4 is able to provide a second signal (e.g., non-antigen specific) for T cell stimulation, such as that seen in transplantation. The monoclonal antibody MK-2 has been used in transplant studies to suppress cardiac rejection and induce long-term cardiac graft survival. In addition to inflammatory responses, VCAM-1 has a significant role in hemopoiesis through its ability to retain lymphocyte and myeloid precursors on stromal cells in the marrow and lymphoid organs. CD106/VCAM-1 exists as an integral membrane protein. The M/K-2 monoclonal antibody immunoprecipitates a peptide that gives a single band on SDS-PAGE gels with an apparent Mr of ~100 kDa under reducing conditions and 92 kDa under non-reducing conditions.

Description:   The monoclonal antibody 14B11 reacts with murine Ly49C, Ly49F, Ly49H, and Ly49I, members of the lectin-like homodimeric Ly49 family of cell surface receptors. Mouse Ly49 receptors exhibit allelic specificity for MHC class I Ia molecules and are thought to serve to prevent natural killer (NK) cells from attacking normal cells while allowing them to attack infected or transformed cells in which class I molecules have been downregulated. These inhibitory receptors are also expressed on a subpopulation of mouse CD8+ T cells. The 14B11 antibody stains ~90% of NK1.1+CD3- splenocytes and ~6% of CD8+ spleen cells from C57BL/6 mice. The antibody also stains 30-80% of NK1.1+CD3- splenocytes in other mouse strains tested (BALB/c, C3H, 129, SJL, AKR/J).

Description:   The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. It is transiently expressed at a low level during B cell development in bone marrow on the CD45R/B220low TdT- sIg- pre-B II and CD45R/B220low TdT- sIgM+sIgD- immature B cells, but not on the CD45R/B220low TdT+ sIg- pro-B/pre-B I stage nor on CD45R/B220high TdT- sIgM+sIgD+ mature B cells. CD25 is expressed at a higher level during very early T-cell development in fetal and adult thymus. The 7D4 monoclonal antibody reacts with an epitope between amino acids 125 and 212 that is distinct from that recognized by the monoclonal antibody 3C7 (SB Cat. No. 1600). While the monoclonal antibody 7D4 does not block the binding of IL-2 to CD25, when used in combination with the monoclonal antibody 3C7 it results in higher levels of inhibition of IL-2-driven proliferation than either of these two monoclonal antibodies alone.

Description:   CD81, also known as TAPA-1, is an integral membrane protein expressed on a variety of cell types and has a high degree of sequence homology between human and mouse. CD81 is expressed on thymic stromal cells where it plays an important role in the transition of γδ+ T cells to more mature T cells with αβ T cell receptors. Immunohistochemical staining has revealed that its expression is localized to the subcapsular region of the thymus and, specifically, on cells that have distinct clustering patterns. It has been speculated that the ligand for CD81 is the pre-T cell receptor which is composed of a TCR β chain and glycoprotein pTα. The monoclonal antibody 2F7 can block thymocyte interaction with CD81 in vitro.
