CD45, also known as the leukocyte common antigen (LCA), exists as four different isoforms (180, 190, 205, 220 kDa) that are generated by alternative splicing of three exons. LCA is found on all cells of hematopoietic origin, except erythrocytes and platelets. CD45 expression is required for T cell and B cell antigen receptor-mediated activation.

The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosporylate the Src family of protein kinases. This activity is known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685 and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody 104 recognizes CD45 (leukocyte common antigen) on all leukocytes of mouse strains expressing the CD45.2 allotype (e.g., A, AKR, BALB/c, CBA/Ca, CBA/J, C3H/He, C57BL, C57BR, C57L, C58, DBA/1, DBA/2, NZB, SWR, 129).

CD11b, the integrin αM subunit, combines with CD18, the integrin β2 subunit, to form the integrin Mac-1, also known as complement receptor 3 (CR3), which mediates adhesion to C3bi and ICAM-1/CD54. Mac-1 is expressed at varying levels on granulocytes, macrophages, dendritic cells, NK cells, and B-1 cells in the peritoneal and pleural cavities. Mac-1 expression is rapidly up-regulated on neutrophils after activation and in the same time period that CD62L is shed from the cell surface. The “classical” MHC Class I molecules are histocompatibility antigens encoded by the H-2 gene complex and consist of heterodimers of highly polymorphic α chains noncovalently associated with the invariant β2-microglobulin. These antigens are expressed on most nucleated cells but expression varies on different cell types. MHC Class I molecules present endogenously synthesized peptides to CD8+ T lymphocytes which are usually cytotoxic T cells. MHC Class I antigens expressed on thymic epithelial cells regulate the positive and negative selection of CD8+ T cells during T cell ontogeny. The monoclonal antibody 28-14-8 binds to the α3 domain of H-2Db in the presence or absence of β2 microglobulin. It cross reacts with the α3 domain of H-2Ld, but not Kd or Dd, and with H-2Dq and/or Lq. Mouse NKG2 is a natural killer (NK) cell receptor for the non-classical MHC class I molecule Qa-1b. NKG2 is expressed on NK cells, lymphokine-activated killer (LAK) T cells, and some CD8+ memory T cells. Two-color immunofluorescent staining with the 20d5 antibody reveals two distinct NKG2+ phenotypes within the NK1.1+CD3- spleen cell population: NKG2hi (50%) and NKG2low (50%). With the exception of DBA/2J mice, NKG2 is expressed on NK1.1+ cells from all mouse strains tested (C57BL/6, BALB/c, 129/J, C3H.SW, AKR/J, SJL).
