CD36, also known as GPIV, is an 88 kDa glycoprotein that is a receptor for extracellular matrix proteins such as collagen and thrombospondin. Its expression is restricted to platelets, monocytes, macrophages, erythrocyte precursors, adipocytes, activated keratinocytes, and some endothelial and epithelial cells. CD36 is a multifunctional protein that has roles as a cell adhesion molecule, a scavenger receptor, a signal transducer and in the pathogenesis of malaria via its affinity for Plasmodium falciparum erythrocyte membrane protein 1 (PEMP1). CD36 on macrophages functions in the phagocytic clearance of apoptotic cells.

In the chicken, the CD8 molecule is present in two forms - (i) a homodimer of two α chains and (ii) a heterodimer of an α chain and a β chain. Chicken CD8 is expressed on approximately 80% of thymocytes, 15% of blood mononuclear cells and 50% of spleen cells but less than 1% of cells in the bursa and bone marrow. While the vast majority of CD8+ cells in the thymus, spleen, and blood of adult chickens express both CD8α- and CD8β-chains, a relatively large proportion of the CD8+ TCRγδ cells in the spleens of embryos and young chicks express only the α-chain of CD8. Among intestinal epithelial lymphocytes, the major CD8+ T cell populations present in mice are conserved but there is a population of TCRγδ CD8αβ cells in the chicken that is not found in rodents. The monoclonal antibody EP72 recognizes the CD8α chain.

The monoclonal antibody TCR-1 precipitates a heterodimer of Mr 90 kDa (two bands of Mr 50 kDa and 40 kDa upon reduction) on chicken peripheral blood T cells. Deglycosylation of the heterodimer yields two polypeptides of Mr 35 kDa and 32 kDa. Unlike human and mouse in which γδ cells comprise a minor subset of T lymphocytes in the circulation, the chicken has a relatively large subset of γδ T cells. The frequency of TCRγδ cells is typically 20-25% of the total blood T cells but may reach approximately 50% in chickens of six- months of age. The majority of TCRγδ+ cells in the thymus and blood are CD4-CD8- although a small subset of them may express CD8 or CD4 coreceptors. However, when the TCRγδ+ cells migrate into the spleen and intestine most of them begin to express CD8. Although the biological function of γδ T cells is unclear, they are clearly capable of cytotoxic activity in vitro. The CD8+ γδ T cells may also be involved in down-regulation of the immune response. They cannot, however, induce a graft-vs-host (GVH) reactions.

The KM201 monoclonal antibody reacts with an epitope very close to the hyaluronate binding domain on the CD44 molecule. KM201 can inhibit hyaluronate-dependent cell aggregation, prevent lympho-hemopoiesis in both Dexter and Whitlock-Witte cultures, prevent the earliest intrathymic precursors from homing to the thymus, and costimulate the activation of freshly purified splenic CD4+ T cells.
