Description:
CD33 is a 67 kDa type I transmembrane glycoprotein and a member of the sialoadhesin family of cell surface receptors. It is absent from pluripotent stem cells but appears on myelomonocytic precursors after CD34. It then continues to be expressed on both the myeloid and monocyte lineages. While it has been reported that CD33 can function as a sialic acid-dependent cell adhesion molecule, cells expressing CD33 require desialylation before they can bind cells bearing the appropriate sialoglycoconjugates. This suggests that inhibitory cis interactions may regulate or block any adhesion function.

Description:
CD107a, also known as lysosomal-associated membrane protein 1 (LAMP-1), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains, and is a ligand for E-selectin-mediated cell adhesion. CD107a is expressed by activated T cells, macrophages, dendritic cells, activated platelets, tonsillar epithelium, and some tumor cell lines, including U937 and KG1a. It is also a widely expressed intracellular antigen. LAMP-1 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases. The upregulation of both CD107a and CD107b on the surface of tumor cell lines has been associated with their enhanced metastatic potential where they may increase adhesion to extracellular matrix and endothelium.

CD19 is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily. It is expressed at all stages of B cell differentiation except terminally differentiated plasma cells. It is also present on the cell surface of follicular dendritic cells. CD19 associates with CD21, CD81, Leu 13 and/or MHC Class II molecules to form a signal transduction complex on the B cell surface. This signaling complex modulates the activation threshold for the B cell antigen receptor (BCR).

CD158e1 is a 70 kDa transmembrane glycoprotein and a killer cell immunoglobulin (Ig)-like receptor (KIR) that is expressed on a subset of natural killer cells and T cells. The monoclonal antibody DX9 is expressed on a subset of CD3-, CD56+ NK cells isolated from peripheral blood at varying levels (<0.1% to 61%) among individuals but not on thymocytes, B lymphocytes, granulocytes, or monocytes. Interactions with CD158e1 and certain MHC HLA-B antigens on a target cell inhibits cytotoxicity, possibly preventing target cell lysis and death. KIR and MHC class I interactions may regulate NK and T cell response to antigen stimulation.

CD49b, a member of the integrin family of extracellular matrix and cell adhesion molecules, is the integrin α2 chain. CD49b associates noncovalently with CD29, the integrin β1 subunit, to form the heterodimer integrin α2β1 that binds laminin and collagen. It is expressed on B and T lymphocytes, NK cells, monocytes, platelets, fibroblasts, and long-term cultured T cells. CD49b/CD29 mediates the Mg++-dependent adhesion of platelets to collagen. It may also be utilized by fibroblasts for the reorganization of the collagen matrix during wound healing.

Description:
The immunoglobulin heavy chain binding protein BiP is a member of the hsp70 family of heat shock proteins and is identical to the glucose regulated protein GRP78. While BiP was originally described for its function in B cells, it is now known to be distributed in a variety of tissues, if not ubiquitous. The highly conserved hsp70 proteins have an essential physiological role in stress responses and as “molecular chaperones” which are responsible for a variety of functions such as protein transport, prevention of protein toxicity and direction of protein folding. With regard to its immunological role, BiP is a component of the endoplasmic reticulum and binds free intracellular heavy chains in nonsecreting pre-B cell lines (μ+,L-) or incompletely assembled Ig precursors in H+L+ secreting hybridomas and myelomas. In the absence of light chain synthesis, heavy chains remain associated with BiP and are not secreted. BiP is an ATP binding protein and the dissociation of the BiP-heavy chain complex is probably driven by the ATPase activity attributed to BiP. The monoclonal antibody 76-E6 recognizes a conserved epitope localized within the region of amino acids 497 to 581 of BiP.

CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow. The monoclonal antibody 6D5 recognizes the same epitope as the published clone, 1D3.
